The main purpose of this study is to explore the low temperature treatment on pig epididymal sperm motility and in vitro fertilization. This test is used mainly CASA, sperm capacitation treatment, in vitro fertilization and other methods of analysis, the following results:1. pig sperm injection, epididymal sperm motility, plasma membrane integrity and acrosome integrity than the test group sperm high.2. pig sperm injection, epididymal sperm VCL (curvilinear motion average velocity), VSL (average linear rate of movement), VAP (average path velocity) than the test group sperm high.3. The pig sperm injection, sperm, low sperm after treatment, after cold shock pig sperm capacitation and mature oocytes were treated in vitro fertilization, the sperm was observed 4-5 hours after oocyte maturation attached conditions. The results showed that:fresh semen than sperm adhesion, adhesion case the cold shock of epididymal sperm is not as cold treated epididymal sperm.4. The pig sperm injection, sperm,-196℃ frozen-thawed sperm,-80 "C frozen-thawed sperm, sperm cold shock after capacitation and mature oocytes in vitro fertilization, the oocytes was observed after 2 days cleavage, and to calculate the rate of cleavage. Sperm injection cleavage rate (60.37 ± 11.7%), sperm cleavage rate (59.34 ± 10.05%),-196℃ frozen-thawed sperm cleavage rate (30.97 ± 5.18%),-80℃ frozen-thawed sperm cleavage rate (24.64 ± 4.32%), cold shock epididymal sperm cleavage rate (10.97 ± 2.18%). The results showed that:injection of sperm and sperm cleavage rate is basically the same,-196℃ frozen-thawed sperm cleavage rate than -80℃ frozen-thawed sperm, sperm cold shock of the lowest rates of cleavage.
