This dissertation consists of two parts:Part 1:The expression levels of S100A8, S100A9, β-Catenin which is Wnt/β-Catenin pathway key protein, and matrix metalloproteinase 7 (MMP7), a target gene of Wnt/β-Catenin pathway and its correlation with clinicopathological characteristics were measured by immunohistochemistry in nasopharyngeal carcinoma (NPC) tissue. Part 2:Effect of the Expression of S100A8 and S100A9 on the Wnt/β-Catenin Pathway was analyzed in NPC CNE1 cell.Part 1:The expression levels of S100A8, S100A9, β-Catenin, MMP7 and its correlation with clinicopathological characteristics were measured by immunohisochemistry in nasopharyngeal carcinoma (NPC) tissue.OBJECTIVE:The expression levels of S100A8, S100A9, β-Catenin, MMP7 in NPC tissue and its correlation with clinicopathological characteristics were investigated.METHOD:1. the positive staining area percentage of S100A8, S100A9, β-catenin and MMP7 were measured by immunohistochemistry in NPC and chronic pharyngitis (CP) tissue. The expression of SI 00 A8 and S100A9 were detected in 49 cases of NPC tissues and 20 cases of non-cancer tissues. The expression of P-Catenin and MMP7 were determined in 42 cases of NPC tissues and 9 cases of non-cancer tissues.2. All data was analyzed by SPSS 16.0 software. Data were expressed as median and interquartile range (P25-P75) and difference between two groups was compared by Mann-Whitney U tests.RESULTS:1. the percentage of S100A8-S100A9-β-Catenin-and MMP7-positive area in NPC tissues was significantly higher compared with chronic nasopharyngitis tissues (P<0.05). The percentage of S100A8-S100A9-β-Catenin-and MMP7-positive area in stage Ⅱ, Ⅲ, and IV NPC was significantly higher compared with chronic nasopharyngitis tissues (P<0.05), and the percentage of positive area in stage Ⅲ, and IV was significantly higher than that in stage II (P <0.05).2. The results of Correlations with the clinicopathological characteristics indicated that the percentage of S100A8-S100A9-β-Catenin-and MMP7-positive area in NPC tissues was correlated with clinical staging (P< 0.05), but nor related to gender or age.CONCLUSION:the expression of S100A8, S100A9, β-Catenin, MMP7 in NPC tissues was up-regulated, which was related to the clinical stage. Terminal NPC was serious than early and medial stages.Part 2:Effect of the Expression of S100A8 and S100A9 on the Wnt/β-Catenin Pathway was analyzed in NPC CNE1 cell.OBJECTIVE:Detection of β-Catenin which is Wnt/β-Catenin pathway key protein, and matrix metalloproteinase 7 (MMP7), a target gene of Wnt/β-Catenin pathway in CNE1 cell cultured using add 1μg/ml S100A8/A9.METHOD:1. Detection of β-Catenin in CNE1 cell cultured using add 1μg/ml S100A8/A9 by the immunocytochemistry and Western Blot.2. Detection of MMP7 in CNE1 cell cultured using add 1μg/ml S100A8/A9 by RT-qPCR.RESULTS:1. It was shown that the number of β-Catenin-positive cells in the experimental group was significantly higher, and the expression of β-Catenin was markedly increased compared with the control group.2.The relative expression level of MMP7 was detected by RT-qPCR. The results indicated that The expression of MMP7 mRNA in the experimental group was significantly higher than that in the control group (P<0.01).CONCLUSION:In vitro cell experiments showed that the ratio of P-Catenin-positive cells and the expression of MMP7 in CNE1 cells cultured in medium containing 1μg/ml S100A8/A9 were significantly increased, suggesting that S100A8, S100A9 might promote the invasion and migration of NPC cells throμgh activating the Wnt/β-Catenin pathway.
