Objective: To study intervention effects of sufentanil(Suf)preconditioning on pirarubicin(THP)-induced acute myocardial injury in rats, and whether to have protective effects. This study will provide many experimental basis for the clinic.Methods: A total of 30 female SD rats, weighing 240-260 g, were randomly divided into three groups with 10 rats each as follows: Group A(glucose saline), group B(THP) and group C(Su+THP).(1) Group A: Same volume of glucose saline with THP were intraperitoneal injection in the first day of experiment.(2) Group B: The first day of the trial a one-time intraperitoneal injection of THP dose of 15mg/kg, injection completed in 2 min.(3) Group C: The first day of the trial one time Suf were intraperitoneal injection with dose of 10ug/kg, completed injection in 2 min; and a one-time THP were intraperitoneal injection with dose of 15mg/kg in 30 min later, and completed injection in 2 min. In the forth day of the trial, the blood sample were taken from abdominal aorta after anesthesia, and to detect the level of serum NT-pro BNP and c Tn I. Rats were all killed after taking blood. Myocardial tissue and right carotid arteries were examined by light microscopy. Production of myocardial tissue homogenate and detection of MDA and SOD.Results:(1) Serum levels of c Tn I, NT-pro BNP: They were compared between three groups, the difference were statistically significant(P<0.05). They were higher in group B than that in group A significantly(P<0.01). They were also higher in group C than that in group A significantly(P<0.05);(2) MDA content: They were compared between three groups, the difference were statistically significant(P<0.05). It was higher in group B than that in group A significantly(P<0.01). It was also higher in group C than that in group A significantly(P<0.05);(3) SOD activity: They were compared between three groups, the difference were statistically significant(P<0.05). It was lower in group B than that in group A significantly(P<0.01). It was also lower in group C than that in group A significantly(P<0.05). It was also lower in group C than that in group B significantly(P<0.05).(4) Myocardial pathological score: They were compared between three groups, the difference were statistically significant(P<0.01). They were higher in group B, group C than that in group A significantly(P<0.01). It was also lower in group C than that in group B significantly(P<0.05).(5) Carotid artery pathology: Group A of normal cell morphology, muscle fibers arranged neatly. Group B of muscle fiber degeneration and focal necrosis, interstitial edema, cell vacuole. Muscle fiber arrangement were normal in Group C, and the interstitial edema, a small amount of vacuoles in the cells, and the cytoplasm eosinophilic change.Conclusion:(1) THP induced myocardial injury and carotid artery injury in rats;(2) Suf preconditioning had a protective effect on THP induced acute myocardial injury in rats.
