| Aim To investigate the effect and mechanism of sijunzi decoction (SJZD) on the intestinal mucosal barrier, we use the murine of dextran sulphate sodium (DSS)-induced ulcerative colitis for this study.Methods Fifty male BALB/c mice of weighting 18g-20g were randomly divided into five groups, including normal group, DSS group, SJZD low dosage group, SJZD medium dosage group and SJZD high dosage group, every group has ten mice. From day 1 to day 7, normal group were given distilled water to drink freely, the other four groups had been given distilled water containing 3% concentration DSS to induced experiment colitis. From day 8 to day14, the normal group and DSS group were given by gavage with distill water. Meanwhile, SJZD low dosage, SJZD medium dosage and SJZD high dosage group were fed by the same way but with different dosage of SJZD(2.25g/kg, 4.50g/kg and 9.01g/kg respectively). Source of disease active index(DAI) were collected everyday, DAI include weight loss, stool consistency and bleeding(occult/gross). On day 15,plasma and colon tissue were obtained from all mice. Levels of plasma D-Lactate were measured by ELISA, and expression of occludin were evaluated by immunohistochemistry.Results 1. The result of DAI and histologic scores:Compared with the normal group, the DAI and histologic scores were significantly elevated in the DSS group and other three SJZD groups(P<0.01). However, DAI and histologic scores were lower in the SJZD medium dosage group and SJZD high dosage group than the DSS group(P<0.01). In addition, there were no significant variance in DAI and histologic scores between the DSS group and SJZD low dosage group(P>0.05).2.The level of Plasma D-Lactate:Plasma D-Lactate was significantly increased in the DSS group (P<0.01)and decreased in other three SJZD groups(P<0.01).3. The expression of occludin:The density mean of DSS group was lower than the normal group(P<0.01). Compared with the DSS group, the expression of occludin of the SJZD medium dosage group(P<0.05) and SJZD high dosage group were improvement(P<0.01). The same as the result of DAI and histologic scores, there were no difference between the DSS group and SJZD low dosage group in the expression of occludin(P>0.05).Conclusion 1. SJZD can ameliorate the clinical manifestations and the microscopic performance of ulcerative colitis in mice induced by DSS.2. Meanwhile, SJZD could decrease plasma D-Lactate so that it improve the permeability of intestinal mucosal barrier 3.SJZD could increase expression of occludin, its mechanism may relate to protect the intestinal mucosal barrier. |
