Objective:The purpose is to evaluate the prognostic value of nm23-H1 expression in adrenocortical carcinoma by investigating the expression of nm23-H1 in the human adrenocortical carcinoma cells (SW-13) subcutaneous transplanted model and peritoneal metastasis model groups.Method:(1) Nude mice were injected with human adrenocortical carcinoma SW-13 cell and randomized into human adrenocortical carcinoma subcutaneous transplanted model, peritoneal metastasis mode model and control groups.(2) HE observation, the expression of Ki-67 by immunohistochemistry and tumor cells primary cultured were used to study the morphology and tumorigenicity.(3) nm23-H1 expression in the human adrenocortical carcinoma subcutaneous transplanted model and peritoneal metastasis model groups was detected by real-time PCR and Western blotting.Results:(1) It was successful to construct subcutaneous transplanted model and peritoneal metastasis model model of human adrenocortical carcinoma cells (SW-13) in nude mice. The tumor formation rate was 100%,80% in nude mice two models respectively.(2) In two constructing microenvironment in vivo, it was easy to catch sight of pathological mitotic figure by HE staining and Ki-67 protein was high expression in subcutaneous and metastatic tumors. Tumor cells primary cultured are similar to SW-13 cells.(3) The expression of nm23-Hl mRNA was 1.235±0.047, 0.765±0.037 in SW-13 subcutaneous transplanted model and peritoneal metastasis model groups respectively; and the nm23-H1 protein level was 1.80±0.07,1.66±0.13,0.81±0.11 in SW-13 cells, SW-13 subcutaneous transplanted model and peritoneal metastasis model groups respectively. Comparison among the two tissues showed that the nm23-H1 mRNA and protein levels of human adrenocortical carcinoma subcutaneous transplanted model group was higher than that of peritoneal metastasis model groups (p<0.05).Conclusion:(1) It was successful to construct subcutaneous transplanted model and peritoneal metastasis model of human SW-13 adrenocortical carcinoma in nude mice. The models suggested that human adrenocortical carcinoma cells (SW-13) in nude mice had strong capacities of tumorigenicity, invasion and metastasis.(2) nm23-Hl expression seems to be a prognostic factor for adrenocortical carcinoma.
