Objective to monitor the background rodent species of jiangsu ports,detect the pathogen,and built the mt DNA-Cytb gene characteristics database of these rodent species.Method in May 2014-April 2015, we collected samples of the common rodent species in 17 ports and identified them by morphological characteristics, then got rat liver and lung through anatomy in a biological safety cabinet, acquired the Cytb gene of rodent species through gene amplification and sequencing,by PCR,compared with GenBank of NCBI by BLAST,took the highest homology as molecular identification results,comparing with morphological identification results. Screened DNA templates of rat liver and RNA templates of rat lung for Yersinia pestis bacteria and Hantavirus by fluorescent PCR, then the positive results were sequenced and typed.At last,combined the Cytb gene orders of the representative samples with geographic distribution information of the common rodent species in 17 ports,built gene characteristics database of the background rodent species of Jiangsu ports,and built the phylogenetic trees of Cytb gene.Result We catched 418 rodent species,including 231 Rattus norvegicus,38 Rattus tanezumi,99 Mus musculus,44 Apodemus agrarius,6 Crocidura lasiura.Molecular identification results and morphological identification results are in complete agreement.All the Screening results for Yersinia pestis bacteria are negative,18 Screening of RNA templates of rat lung for Hantavirus are positive,4.31% of all the screening results.In all the18 positive results,15 types are SEO type,the host of which is Rattus norvegicus of Lianyungang ports,the left 3 types are HTN,the host of which is Apodemus agrarius of Nanjing Longtan Port. Built Gene characteristics database of the background rodent species of Jiangsu ports,in which Nantong, Yangzhou, Nanjing, Zhangjiagang,Wuxi,Zhenjiang, Lianyungang,Huaian have the representative samples of Apodemus agrarius,Nanjing Zhangjiagang Zhenjiang Jiangyin Changzhou Xuzhou Taizhou ChangshuSuqian have the representative samples of Mus musculus,Wuxi Lianyungang,Suqian,Taicang have the representative samples of Rattus norvegicus,Yangzhou Nanjing Zhangjiagang Taizhou Taicang Wujiang Kunshan Suzhou have the representative samples of Rattus tanezumi. The phylogenetic trees of Cytb gene established by gene characteristics database can clearly distinguish various rodents.Combine the gene characteristics database of the above rodent species and reference rodent species,then form the phylogenetic tree of 5 independent rodent species.Conclusion Common rodent species in 17 ports of Jiangsu are Apodemus agrarius,Mus musculus, Rattus norvegicus and Rattus tanezumi, accidental species is Crocidura lasiura while other rodent species are not found. Molecular identification results and morphological identification results are in complete agreement.The molecular identification can meet the actual needs of the identification of rodent species in ports.Yersinia pestis bacteria are not detected, while Hantavirus are detected in Rattus norvegicus of Lianyungang Port and Apodemus agrarius of Nanjing Longtan Port.There is a risk of occurrence and prevalence for epidemic hemorrhagic disease, the sanitary treatment and monitoring of which should be enhanced. The risk evaluation based on the phylogenetic trees of Cytb gene of main rodent species in Jiangsu ports show that there is a high risk of pandemic of type Ⅰ hemorrhagic fever with renal syndrome in Nanjing,Wuxi,Nantong,Yangzhou and Lianyungang ports,comparing with a high risk of pandemic of type Ⅱ hemorrhagic fever with renal syndrome in Lianyungang and Suqian.
