Objective:To investigate the association of serum LIF level with obesity and type 2 diabetic mellitus. and to analyze the correlation between LIF and the age, the parameters of body fat, glucose, lipid metabolism, and insulin sensitivity, to explore the role of LIF in obesity and type 2 diabetes pathogenesis and development.Methods:On the basis of body mass index(BMI),44 newly diagnosed T2 DM patients(T2DM group) were divided into groups of overweight and obesity group,( A group,22 cases,BMI≥25 and B group, 22 cases, 18.5kg/m2≤BMI<25 kg/m2), 44 subjects with normal glucose tolerance(NGT group) were divided into groups of overweight and obesity group,( C group, 22 cases,BMI≥25 and D group, 22 cases, 18.5kg/m2≤BMI<25 kg/m2). Height, weight, Blood pressure, waist circumference(WC),Hip circumference(HC), total cholesterol(TC), triglycerides(TG), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol(HDL-C), fasting plasma glucose,glycosylated hemoglobin(Hb A1C), C-reactive protein et al were determined in all subjects. Body mass index(BMI), waist-to-hip ratio(WHR) and homeostasis model assessment for insulin resistance index(HOMA-IR) were calculated. The level of serum LIF was determined by ELISA.Results:The serum level of LIF was higer in T2 DM group than that in NGT group(P<0.01),which was higer in A group than that in groups of C and D(P<0.01). The level of FBG and Hb A1 c and LIF was higer in A and B groups than that in groups of C and D(P<0.05). The level of CRP and IL-6 in A group was higer than that in groups of C and D(P<0.05). The level of FBG, IL-6, CRP, LIF and BMI was higer in A group than that in group of B(P<0.05). Meanwhile The level of LIF, IL-6, BMI, CRP, TG in C group was higher than that in D group(P<0.05). The serum level of LIF was positively correlated with Age、FBG、Hb A1c、CRP、BMI、waist-hip ratio and IL-6(r=0.228,0.729, 0.665,0.852, 0.627,0.601,0.911,P<0.01), Muitiple regression analysis showed that CRP and IL-6 was the independent influencing factor of serum LIF level(β=0.317,0.753,P<0.01).Conclusion:(1) The serum level of LIF in type 2 diabetes patients was significantly higher than that of the normal control group, diabetes of overweight and obesity group was higher than those two sub groups of the non diabetes mellitus group.These suggest that elevated LIF level may promote the occurrence and development of obesity and type 2 diabetes mellitus.(2) LIF was positively correlated with Age、BMI、FBG、Hb A1c、waist-hip ratio and CRP、IL-6,so it may be delay the progress of diabetic course when we have a good controlling of fasting blood glucose and glycosylated hemoglobin, weight loss, meanwhile can decrease the level of LIF,(3) CRP and IL-6 are independent factor of LIF, this suggesittng that LIF may be involved in inflammation. The LIF is close to IL-6 which were worked with each other to promote the pathogenesis and development of T2 DM and obesity.
