Objective: To detect the expression of Plexin-B1 and MMP-9 in tissues of normal pancreatic and pancreatic carcinama, in order to analyze the correlation of the expressions and the relationship between Plexin-B1, MMP-9 and clinical pathological factors, follow up the patients with pancreatic cancer after surgery in order to get the survival time, explore the significant of the expressions of Plexin-B1 and MMP-9, and the related factors that influence the survival time after operation of patients with pancreatic carcinoma.Method:1. 39 specimens from patients who were diagnosised with pancreatic carcinoma and received operation in Hunan Provincial People’s Hospital from January 2012 to December 2015 were put in the study. The 39 cases were staged according to the 7th TNM staging system of the American Joint Committee On Cancer. Get the patients’ clinical pathology data by consulting the basic medical records including age, gender, degree of differentiation and etc. Another 10 specimens of normal pancreatic tissues from patients with pancreas trauma and received pancreatic resection were chosen as control group. To detect the expressions of Plexin-B1 and MMP-9 through immunohistochemical staining, and compare the differences between normal pancreatic tissue and pancreatic cancer tissue. To observe the neural invasion phenomenon in pancreatic cancer tissue through HE staining. And then, analysis the relationship between the expressions and clinical pathologic data and correlation of expressions.2. The survial time were calculated from the time of operation to the death of the patients or the last follow-up until December 2015. The data of survial time were obtained through E-mail, out-patient or hospital review, SMS, telephone and ect. The expressions of Plexin-B1 and MMP-9 and the clinical pathology data were taken into univariate analysis of survial time. After univariate analysis, the factors that might influence the survial time(P<0.1) were included in the multivariate analysis.Results:1. As the immunohistochenmical result show, Plexin-B1 and MMP-9 were mainly expressed in pancreatic cancer cell cytoplasm and cell membrane, both of them showed a trend of high expression(97.4% and 92.3%), significantly higher than the normal pancreatic tissues(10% and 10%), the differences were statistically significant. The expression of MMP-9 is higher in pancreatic cancer tissue with nerve invasion and low degree of tumor differentiation, the differences were statistically significant. The expressions of Plexin-B1 and MMP-9 in pancreatic cancer tissuess were positvely correlated relationship(r=0.483, P=0.002).2. Excluded the 2 patients died of postoperative complications, the remaining 32 patients’ clinical pathology factors were taken into the univariate analysis of survival time. The results showed that the over-expression of MMP-9, nerve invasion, low degree of differentiation and later TNM staging were associated with shorten survival time(P < 0.05). The factors that might influence the survial time(P < 0.1) were included in the multivariate analysis of Cox model, results suggest that neural invasion and low degree of tumor differentiation were relatively independent factor of shorten postoperative survival time, relative risks were 0.215(P=0.002) and 3.207(P=0.02).Conclusion:1.Plexin-B1 and MMP-9 were highly expressed in tissue of pancreatic carcinama, higher than normal pancreatic tissues.2. The expressions of Plexin-B1 and MMP-9 in pancreatic carcinama were positvely correlated relationship, suggest that Plexin-B1 and MMP-9 were correlated with the pancreatic carcinoma’s tumorigenesis and development process.3. The survival analysis suggests that over-expression of MMP-9, nerve invasion, low degree of differentiation and later TNM staging were remarkably associated with shorten survival time, neural invasion and low degree of tumor differentiation were relatively independent factor of shorten postoperative survival time.
