Objective:To investigate Th17 cell-associated factor IL-17 and mTOR signaling pathway of the target proteins p-S6 protein that express in LN kidney tissues, and to explore whether the mTOR signaling pathway is activated in the kidney tissues of patients with LN, correlation among types (Ⅱ, Ⅲ, Ⅳ, Ⅵ), the level of activity and pathogenesis of LN. Hence, this can provide a new target for patients with LN.Methods:A total of 35 cases of kidney tissue paraffin blocks were collected from the Second Affiliated Hospital of Kunming Medical University, Dermatology/Rheumatology department. According to SLEDAI score and renal activity index, the patients were divided into peripheral stable group/peripheral activity group(SLEDAI<6, SLEDAI>6) and low activity group renal group/high activity renal group(AI≤6, AI>6). Among them 3 cases of glomerulonephritis patients with kidney tissue paraffin blocks were taken into control group irrespective of infection and other autoimmune diseases. At first immunohistochemical staining was performed in kidney tissues of 13 LN patients (case group) and glomerulonephritis tissues (positive control group) that detected IL-17 and p-S6. Then immunohistochemical double staining was performed that detected IL-17 and p-S6 at the same time. The expression level of p-S6 and IL-17 was observed under ordinary light microscope by microscopic imaging software to capture images respectively. Their amounts were differentiated with an IPP software, which gave the results of integrated optical density values. Using SPSS 17.0 software package for statistical data analysis, data are expressed x±S. Between the two groups were compared by using two independent samples t-test. A pair wise comparisons LSD-t test, correlation analysis was done using Pearson & Spearman correlation analysis, while P<0.05 showed a statistically significant difference.Results:l.In LN renal group IL-17 and p-S6 are 4311.64 ±3082.11,577.06±399.82 respectively. The IL-17 and p-S6 from LN patient renal tissue showed a positive correlation between the expression levels(r=0.800, P<0.01).2.Peripheral activity:In peripheral activity group and stable group urine protein levels in the two groups are 2.17±1.88g/24h,2.24±2.41g/24h respectively. Corticosteroid dosage are 61.60±25.55mg/d,45.65±32.56mg/d respectively. p-S6 integral optical density are 494.97±374.18,886.34±405.06. The p-S6 expression of LN in kidney was peripheral activity group less than stable group(P<0.05).3.The renal pathological activity index (AI,8.450±4.677) calculated by Pearson method, by single factor analysis, there was a negative correlation between p-S6 expression and renal pathological activity index (r=-0.432, P<0.05). Renal group high and low activity protein levels are 3.11±2.13g/24h,1.25±1.24g/24h respectively. Corticosteroid dosage are 71.27±26.64mg/d,52.00±32.29mg/d respectively. p-S6 integral optical density are 518.50±406.63,878.52±366.75 respectively. Intrarenal high activity group p-S6 expression levels below the low activity group (P<0.05).4.Glucocorticoid:Glucocorticoid dose average of 22 patients with LN 65.71±32.01 calculated by Pearson method, by single factor analysis, there was a negative correlation between p-S6/IL-17 expression and glucocorticoid dose average (r=-0.534, P<0.05, r=-0.722, P<0.05,respectively.).5.Renal pathological classification:In LN patient pathological staging p-S6 expression level is different or as a whole (P<0.01), grade Ⅲ>grade Ⅵ, grade Ⅲ>grade Ⅳ(P<0.05). The rest had no statistically significant on comparison. 6.ds-DNA:p-S6 and IL-17 expression levels and Spearman correlation analysis, it can’t be considered that the expression levels of p-S6/IL-17 and ds-DNA have correlation (rs=-0.001, P>0.05, rs=-0.593, P>0.05, respectively.).Conclusion:LmTOR signaling pathway is activated in LN patients. Th17 cells may be involved in the pathogenesis of LN through mTOR signaling pathway.2.1n comparison with peripheral activity group, the expression of p-S6 increased in stable group of LN patients. Similar comparison with Renal group high activity showed expression of p-S6 increased in the lower score LN patients. Hence, rapamycin, a mTOR signaling pathway inhibitor is significant in the treatment for the patients who have peripheral stable of LN without remission,.3.The expression of p-S6 and IL-17 in renal tissue of LN patients was negatively correlated with glucocorticoid dosage.
