Cytokines Lineage Screening Correlation Between Optic Neuritis Combined Immune Abnormalities

[Objective] Use with ELISA double antibody sandwich method for the principle of cytokine antibody array technologies, combined screening of immune abnormalities in patients with neuritis apparent correlation between cytokine lineage, expression analysis statistically significant cytokine in the development of the disease and changes in meaning, in order to elucidate the molecular level, as the neuritic immunomodulatory mechanism. Looking for biological markers of disease activity or a new target for treatment of the disease, provide new ideas and methods for clinical diagnosis and treatment.[Methods] Taken 22 cases Serum samples, according to their clinical medical records are divided into three groups, depending on the combined immune abnormalities neuritis patients (Y group),13 cases of optic neuritis patients with normal immune group (O group) 7 cases, normal the control group (N) in 2 cases. In using ELISA double antibody sandwich method for the principle of cytokine antibody array technology, serum samples of 22 patients with 40 kinds of screening assay of cytokines. Form a solid phase antibodies specific antibody binding to a solid support, and then the serum to be tested in the corresponding antigen to form immune complexes, washed plus enzyme-labeled antibody, antigen and immune complexes to form HRP-antigen-antibody complex solid phase, i.e., the "sandwich" structure, and finally adding chromogenic substrate, using a laser scanner InnoScan 300 scanning signal (frequency excitation= 532nm), which determines the amount of expression. Using SPSS19.0 version, GraphPad Prism5.0 version of the data analysis software for statistical analysis.[Results] Take the average of N sets of data N1, then the data were taken 13 N1 ratio Y group (abbreviated as Y/N), O group 7 data N1 ratio (abbreviated as O/N), using GraphPad Prism50.01 data analysis software to draw the line box showing the relationship between the Y group is a multiple comparison of the data group N, O group comparison groupN,95% confidence interval, following results were obtained: inY/N, IL-16, IL-Ira, MIP-la, Eotaxin-2, IL-8, TNFb, Eotaxin, IFNg, GM-CSF, IL-6, ICAM-1, TNFa, IL-2, IL-7, IL-10 these 15 kinds of factors the expression level was significantly up-regulated, multiple values are greater than 1.5 times the level of expression of 1-309 showed a downward trend, a multiple of less than 0.5 times; in O/ N, IL-16, IL-lra, MIP-la, Eotaxin-2, IL-8, Eotaxin, IL-6, IL-5, IL-13 expression level of these nine factors was significantly up-regulated, multiple values were more than 1.5 times, G-CSF, MCSF,I-309 three factor expression levels showed a downward trend, it is less than a multiple of 0.5 times. Y group, group O, N set of three pairwise comparison, the use of statistical analysis software SPSS 19.0 homogeneity of variance test and T-test, the following results:Y group and the comparison group O, cytokine expression levels of IL-lra significantly increased, statistically significant difference, homogeneity of variance, P=0.016<0.05; Y group and N group compared the expression levels of the cytokine IL-lra was significantly increased, statistically significant difference, homogeneity of variance, P= 0.014 <0.05; O group and N group compared cytokine TNF RII expression was significantly down-regulated, there are statistically significant differences, homogeneity of variance, P= 0.030<0.05.[Conclusions] The combined immune abnormalities in patients with visual neuritis interleukin-1 receptor antagonist (IL-lra) expression levels were significantly higher than normal immune index and normal patients with optic neuritis; optic neuritis serum tumor necrosis factor receptor 2 (TNFRII) expression levels were significantly lower than normal; combined immune abnormalities in patients with optic nerve and immune index in normal patients with optic neuritis most relevant cytokine expression levels that are statistically no difference.
