[Objectives]:To investigate the effects of total yunnanensis saponin on salivary adenoid cystic carcinoma ACC-LM cells and xengraft model, study its mechanism and in order to provide theoretical basis for clinical treatment of salivary adenoid cystic carcinoma.[Methods]:To observe the morphology of ACC-LM cells following yunnanensis total saponins intervention through microscopic technique and immunocytochemistry and to assay the expression of RSF-1,PI3K and AKT in ACC-LM cells.24 BALB/C mices were divided into three groups:group Ⅰ (normal),group Ⅱ (DMSO),groupⅢ (DMSO+total yunnanensis saponins). Drug intervention when the xengraft model in vivo was established two weeks and record the weight and tumor volume 3 days/times to make the tumor growth curve and compare tumor growth status.The mices were sacrificed four weeks later, stripped the tumor, weighed the tumor to calculate the inhibition rate.The tumor tissue to stained with Hematoxylin-Eosin staining (HE staining), TUNEL apoptosis detection assay and then analysis apoptosis rate and the expression of RSF-1, PI3K and AKT.[Results]:The control group normal cell morphology rule.round and yunnanensis total saponins group shows irregular contour of the large.Immunocytochemical result showed that RSF-1,PI3K and AKT expressed in ACC-LM cells and mainly localized in the cytoplasm.the average gray value of two groups expression of dosing (132.60± 6.66/155.6±3.58、130.80±2.86/161.40±2.97、130.60±2.70/159.40±5.32)there are significant difference (P<0.01).The tumor formation at the 6th day, the success rate is 100%. Growing weight of mice in the group Ⅰ,but group Ⅱand group Ⅲ had no significant change law.There was no difference between Group Ⅰ and Group Ⅱ (P>0.05)about tumor weight. While,Compared to group Ⅲ there are significant differences(P<0.01)among the three groups.At experiment terminal group Ⅰ,group Ⅱ,group Ⅲ tumor weight was 1.79±0.26g,1.63±0.45g,0.85±0.16g, the tumor inhibition rate about group Ⅱ,group Ⅲ was8.93%,52.51% respectively.HE staining group Ⅰ, group Ⅱ tightly packed cells and group Ⅲ cells arranged in loose and more interstitial. Apoptosis rate among the three groups were 8.13%,8.63%,46.27% respectively. RSF-1 expression among the three groups was 112.08±1.78, 113.60±3.60 and 134.65±4.66 respectively. PI3K was 109.53±4.66,109.48±2.67 and 126.83±5.82 respectively. AKT was 106.60±2.19,106.78±1.75 and 127.13±2.73 respectively. There are significant differences (P<0.01).[Conclusions]:Total yunnanensis saponins can inhibit the proliferation of salivary adenoid cystic carcinoma ACC-LM cell in vitro and reduce the expression of RSF-1,PI3K and AKT.Total yunnanensis saponins can inhibit the tumor growth of ACC-LM cell in vivo reduce the expression of RSF-1,PI3K and AKT. RSF-1, P3K and AKT may play an important role in the progress of ACC.
