Objective:Utilizing gene chips to detect common gene expression and differential gene expression in esophageal cancer tissues, corresponding adjacent tissues, metastatic lymph nodes, lung squamous cell carcinoma and corresponding adjacent cancer tissue. Seeking characteristic or relative characteristic explore gene expression profiles in esophageal cancer and exploring the clinical significance of gene expression on pathological diagnosis and differential diagnosis in esophageal squamous cell carcinoma.Methods:Specimens from the First Affiliated Hospital of Kunming Medical surgical resection Intraoperative which pathological diagnose with intraoperative frozen and Postoperative. There are 6 cases of esophageal cancer,6 cases of corresponding adjacent tissues,2 cases of lymph node metastasis,6 cases of squamous lung cancer and 6 cases of corresponding adjacent tissues. Extract RNA with Trzol from corresponding organization, synthesize cDNA probes and then hybridize to a microarray containing 41000 genes (0ligo 44K Microarray), screening differentially expressed genes that expressed multiple≥3 times and P-value≤0.05.Results:Compare Esophageal cancer to corresponding adjacent tissues,there are 5642 differentially expressed genes, containing 2701 up-regulated genes and 2941 downregulated genes; up-regulated genes include MAGEA1, MAGEA6, KREMEN2, MMP12, MAGEA2B, MMP13, MMP1, HMGA2, KRT17, PPP1R14C, VGF, MTL5, MAGEA8, FOXL2NB, DLX5, KRT14, IRF6, WDR72, etc. downregulated genes include ATP1A2、SCARA5、ANGPTL1、ACTG2、PDZRN4、OGN、MAL、CYP3A5、 PLN、SLC2A4、SLITRK3、PDZRN4、ACTA1、HLF、SORBS2、NCAM1、 DES、RYR2、BPIFB1、PRUNE2、SCRG1、KCNB1、MYZAP、ADH1B,etc. Compare Lymph node metastasis tissue to cancer adjacent tissues,there are 415 differentially expressed genes, containing 157 upregulated genes and 258 downregulated genes; up-regulated genes include HMGA2、IGLL1、TNFRSF17、COL10A1、FCRL5、 MGAT5B、LOC283028、BLK、IGJ、RUNX1、PBX4、ESM1、ZNF761、BEND4、 LAMAl,etc. downregulated genes include CAS2、DES、PGM5、ATP1A2、 ADAMTS8、S100P、PLN、IL1RN、MPP7、KBTBD、LDB3、ECM1、 SYNPO2,etc. Compare esophageal squamous cell carcinoma and its lymph node metastasis tissue, there are 550 common expressed genes, containing 247 up-regulated genes and 303 downregulated genes; up-regulated genes include HMGA2、COL10A1、LOC283028、RUNX1、ESM1、ZNF761、TRAF2、CPNE7、 ANKMY1、UBE2C、KNTC1、PABPC1L、HIST1H4K、KCNK12、CCT6A、 KRT18,etc.downregulated genes include CASQ2、DES、PGM5、ATP1A2、 ADAMTS8、S100P、PLN、MPP7、KBTBD12、LDB3、ECM1、SYNPO2、SLPI、 RYR2, etc.Compare Lung squamous cell carcinoma to corresponding adjacent tissues,there are 1155 differentially expressed genes, containing 355 up-regulated genes and 800 downregulated genes; up-regulated genes include MMP12、SPP1、 NTRK2、DLX6、UCHL1、ADAM23、STRA6、GREM1、MMP11、KRT(14,16,17), etc. downregulated genes include BPIFB1, PIP, FOLR1, ANXA3, KRT (13,24), etc.Comparison of esophageal squamous cell carcinoma and Lung squamous cell carcinoma,discovering 143 specific or relatively specific differentially expressed genes,of which 34 genes expression increased,109 genes expression decreased; up-regulated genes include RUNX1、SEZ6L2、PYCR1、HIST1H2BD、HIST1H4K、 PTPRK、MCM5、PABPC1L、SPHK1、CCT6A、TRAF2、KRT18、TRIM78P、 GIGYF1、SNX5、KCNK12, etc. downregulated genes include MPP7、ANKRD37、 LOC101927718、ATP2B4、PER3、ABLIM1、PRKCA、SOGA3、CAV1、TP53I3、 NT5DC3、S1PR3、RELN、CDO1、ADAMTS8,etc.Conclusions:There are 5642 differentially expressed genes exist in esophageal squamous cell carcinoma and corresponding adjacent tissues,415 differentially expressed genes exist in lymph node metastasis and corresponding adjacent tissues. There are common expressed genes 550 in result of esophageal squamous cell carcinoma and corresponding adjacent tissues compare to lymph node metastases and corresponding adjacent tissues; and there are 1155 differentially expressed genes exist in lung squamous cell carcinoma and corresponding adjacent tissues; Compare esophageal squamous cell to lung squamous cell carcinoma, obtaining 143 differentially expressed genes in esophageal squamous cell carcinoma,6657 differentially expressed genes in lung squamous cell carcinoma.According to GO analysis, suggesting that esophageal squamous cell carcinoma have 550 specific or relatively specific expressed genes, which shows it can be indicators in the pathological diagnosis of esophageal cancer, but also can be used as an objective basis for diagnosis of metastatic esophageal squamous cell carcinoma, providing a new idea for discussion of unknown primary tumor site in metastatic cancer. There are 143 and 6567 characteristic or relative characteristic differentially expressed genes in esophageal squamous cell carcinoma and squamous cell carcinoma of the lung, also suggesting it as a reference index for differential diagnosis of esophageal squamous cell carcinoma and lung squamous cell carcinoma,which can be useful attempt for pathological diagnosis and differential diagnosis.
