ObjectiveLung cancer is one of the most malignant diseases. The study on its carcinogenesis, diagnosis and treatment is the hot spots in the current research. Recent studies have indicated that proteases and protease inhibitors are involved in a variety of pathophysiological processes, including the development of cancer, and a growing number of studies have indicated that serine protease inhibitors have a close relationship with tumor. This study intends to elucidate the expression of SPINK5 in non-small-cell lung cancer and normal lung tissues, intends to discuss its staining intensity correlated with non-small cell lung cancer (NSCLC). We also discuss the expression of SPINK5 in NSCLC between it’s clinical and pathological parameters, for the further study of SPINK5 in non-small cell lung cancer (NSCLC) about mechanism of carcinogenesis and it’s clinical application value, this will provide experimental basis to further study SPINK5 and lung cancers.MethodsTo collect 129 cases of non-small-cell lung cancer patients underwent radical surgical resection with cancer tissues and cancer adjacent normal lung tissues in 2009 January to 2015 June in Yunnan tumor hospital. Through the organization and making pathological sections embedded in paraffin, using immunohistochemical staining method, We determined the SPINK5 expression in lung cancer tissue and normal lung tissue expression location and expression intensity, and statistics of its correlation with clinical pathological parameters was analysed.Results1. Among 129 cases of the cancer tissues and adjacent normal tissues HE staining results were good, in accordance with the quality control standards. SPINK5 was expressed in non-small-cell lung cancer and normal lung tissues, and its expression was located in cytoplasm, cell membrane and extracellular.2. SPINK5 was highly expressed in non-small-cell lung cancers compared with adjacent normal lung tissues with statistical significance (66.67% VS.30.23%, p< 0.001).3. The expression of SPINK5 in the lymph node metastasis of NSCLC was increased (76.92% VS.56.25%,P=0.013), which may be involved in the transfer process of NSCLC.4. Stratified analysis showed that SPINK5 was mainly expressed in lung adenocarcinoma patients with lymph node metastasis, the positive expression rate was 82.93%, the difference was statistically significance (p= 0.012).ConclusionThe high expression of SPINK5 may play an important role in the occurrence and lymph node metastasis of non-small-cell lung cancer.
