| Sjogren’s syndrome (SS) is a chronic autoimmune disease that invades the exocrine glands,especially salivary and lachrymal glands.According to clinical experience, professor IN Shi,a famous TCM physician of Jiangsu,create Shengjin Renzao Prescription to treat sjogren’s syndrome,which has obtained good results in clinic.This essay further studies the mechanism of Chinese medical therapy on SS by animal experiments,which provides powerful evidences for clinical applications.Objectives:To detect the immune induced Sjogren’s syndrome model rat’s CD4+CD25+Foxp3T cells (Treg) and Th17 cells level from peripheral blood, at the same time observe the the improvement of the affected glands and the changes of peripheral blood lymphocyte subsets.Methods:The mice in all the groups except the blank group were established the sjogren’s syndrome in mice model by the immune inducing and each group was intervened respectively.The level of CD4+ã€CD8+ã€CD4+ã€CD25+Foxp3T cells and Th17 cells in the peripheral blood of mice was detected by using flow cytometry,and saliva,tear secretion of model rats was determined; At the end of its treatment,observe the change of submaxillary gland tissue morphology.Results:1.During the dynamic observation of the experiments, the secretion of saliva and tear in mice of the model group were remarkably lower than those in the normal group from the first week after modeling, indicating that the dry mouth and dry eye symptoms of the SS model mice were significant. The levels of CD4+T lymphocytes and Th17 cells increased, while that of the CD8+T lymphocytes decreased, and that of the Treg cells showed no evident difference. Pathologic analysis of the submaxillary gland discovered that the HE staining of the submaxillary gland of mice in the model group showed lymphocyte infiltration in the mesenchyme of the submaxillary gland, without any degeneration or necrosis in the acinar epithelium or in the duct.2.All medication groups of the prescription for producing saliva and moistening dryness showed improved secretion of saliva and tear in mice, and the time needed for relieving the symptoms of dry mouth and dry eye varied by different dosages. The prescription for producing saliva and moistening dryness could reduce the level of CD4+T lymphocytes, but increase that of the CD8+T lymphocytes of mice in the SS model group; and could notably decrease the level of Th17 lymphocytes while increase that of Treg lymphocytes of mice in the SS model group. No lymphocyte infiltration in the mesenchyme of the submaxillary gland of mice, and no pathologic changes in the acinar epithelium or duct in all medication groups were found through pathologic analysis of the submaxillary gland.Conclusion:The mechanism of action of the prescription for producing saliva and moistening dryness might be associated with the T lymphocyte response, which played a role in the immunoregulation, and protecting the gland from inflammatory infiltration through reducing CD4+T and Th17 cells levels while increasing CD8+T and Treg cells levels, thus relieving the symptoms. |
