The Study Of The Correlation Of LVH And AS Based On Essential Hypertension And The Influence Of Related Genes About Compound Spanishneedles Particles

Qbjective:To study the abnormal expression of LVH and AS related genes in hypertension and the intervention effect of compound spanishneedles particles.Methods:The rats were divided into three groups, spontaneously hypertensive （SHR） rats group, administration group （SHR+YAO） and control group （WKY）.After eight weeks,the rats were executed and removed hearts.And then wo got the myocardial tissues.Total RNA was extracted from three groups of rats respectively by using TRIzol.After purified, three groups of differentially expressed genes and lncRNAs were obtained to the Affymetrix GeneChip Rat Gene 2.0 ST Array, and then we texted it.Results: ①Through the GO analysis, we got some differentially expressed genes and lncRNAs associated with hypertension,including Vcam1、Tnfsf10、Tgfb2、Ifit1、Irf7、Mtla、 GAS5、SNHG6、NR₀38078、Bs、XR₁46410. Then we analyzed the function of these genes, including immune response, inflammatory response,cell cycle, signal transduction,etc.After the treatment of compound spanishneedles particles, the expression of differentially expressed genes had been changed, from rise to fall.（2）Through the Pathway analysis, we got the most prominent Pathways, including NF-κB signaling pathway、hypertrophic cardiomyopathy （HCM）、arrhythmogenic right ventricular cardiomyopathy (ARVC、MAPK signaling pathway、 dilated cardiomyopathy、apoptosis, etc,and some signaling pathways related to immune response, containing chemokine signaling pathway、T cell receptor signaling pathway and so on.Conclusion:① mRNAs and lncRNAs were involved in the course of hypertension and played a significant role in the development of LVH and AS based on hypertension.②Signal transduction pathways were involved in the process of gene regulation about hypertension.③Compound spanishneedles particles may play a intervention role in the progress of hypertension and its complications,by regulating and controlling the related genes in hypertension,such as Vcaml、Tnfsf10、Tgfb2、Ifitl, etc.④Immune and inflammatory response occupied an important position in the course of the hypertension.
