Objective:To observe the effects of different doses of Yangyin Runchang Formula containing serum on in vitro cultured mouse jejunum and ileum Interstitial Cells of Cajal, through the detection of interstitial cells of Cajal apoptosis and cell cycle regulation, to explore the interstitial cells of Cajal through action mechanism research.Method:Randomly selected 20 BALB/c mice were divided into four groups, the original generation separation extraction and each mouse jejunum and ileum tissue culture, through the cell flow type sorting mature interstitial cells of Cajal, ahead of the prepared rat normal serum and different concentrations of Yangyin Runchang Formula containing serum on interstitial cells of Cajal of group intervention, respectively high dose group, middle dose group, low dose group and blank control group, intervention after 2 days observation interstitial of Cajal cell number and morphology change, and by flow cytometry technical evaluation of Cajal stromal cells apoptosis and cell cycle regulation.Result:Each culture cells obtained by corresponding intervention and observed under the microscope after high statistical, China, the average count of low and normal dose group were respectively (13.94 2.64+; 10.17+1.33; 817+1.47; 6.33+1.63), the number of high dose and middle dose group were compared with the normal group, the expression was elevated (P < 0.01), low dose group and normal group expression without significant difference (P> 0.05) and low dose group and high dose group exist significant difference (P< 0.01), middle dose group and high dose group and low dose group no significant difference (P> 0.05). In apoptosis, as early as the sum of apoptosis and late apoptosis for total apoptosis rate, the blank group, nourishing Runchang decoction containing serum high, low dose group of early and late apoptotic and 16.9%,9.5%,12.4% and 12.6%, total apoptosis rate:blank group> low dose group> dose group> high dose group. Yin Runchang prescription can effectively reduce serum Cajal cell apoptosis. Cell cycle regulation, the blank group, nourishing Runchang decoction containing serum high, medium and low dose group in S phase and G2 phase cells sum accounts for the proportion to be 36.52%,41.02%,43.86%,82.40% respectively, medicated serum group is in the copy number of ICC activities accounted for the proportion of high dose group> middle dose group> low dose group> blank dose group.Conclusion:Yin Runchang Fang can promote the growth of the number of ICC, and can effectively reduce number of Cajal cells apoptosis and enhance cell division ability and activity, which then mechanism may be through the promotion of the ICC number growth and reduce the apoptosis of to promote gastrointestinal motility, for.the treatment of slow transit constipation to seek a new way.
