The Effects Of Danshao Granula Ⅲ On Nephrin,P-nephrin, Arp2/3 In Rats With HSPN

Henoch-Schonlein purpura nephritis is characterized with different degrees of hematuria, proteinuria, skin purpura,urinary tube and edema. And it is the childhood common secondary kidney disease.There is no disease of allergic purpura nephritis in traditional Chinese medicine.According to its clinical manifestations,it belongs to "blood" and "spotted", "hematuria", "edema" category in traditional Chinese medicine. The incidence of HSPN in children is increasing year by year,becoming one of the main secondary kidney disease seriously endangering children’s health. Western medicine is based on anticoagulant, hormone and immune inhibitors.But it exists side effects,poor treatment and other issues.Traditional Chinese medicine with clinical syndrome differentiation and the overall concept as the instruction and Danshao Granula Ⅲ which is based on cooling blood and removing stasis had achieved good results in HSPN.Podocyte-associated protein such as nephirn,p-nephrin,arp2/3 is associated with proteinuria in HSPN.The machanism of action of Danshao Granula Ⅲ has not been studied.Objective:To explore the effects of Danshao Granula 111 on Nephrin,p-nephrin,Arp2/3 in Rats With HSPN so that we can make it clear than how Danshao Granula Ⅲ works it out.Method1.Selected 25 healthy male SD rats whose weight is about 120g-150g and gave them adaptive feeding 1 week.Rats were randomly divided into blank group, model group and Danshao Granula Ⅲ in high dose group, Danshao Granula Ⅲ in moderate dose group, Danshao particles in low dose group, and each group had five.rats.The method of BSA+LPS+ CC14 was used to establish IgA nephropathy model.Dried ginger water was used to establish blood stasis and heat syndrome model,both of which compose the animal model of HSPN.2. At the end of the molding experiment,all the Danshao groups were treated by Danshao Granula Ⅲ.To observe the glomerular mesangial changes under light microscope.The protein expression of nephrin,p-nephrin, and arp2/3 were examined by western blot.3.The data were statistically analyzed with SPSS 19.0 software.Results1.The model rats showed obvious pathological changes in the renal tissues.The pathological changes in the renal tissues of the Danshao groups were reduced in comparison with those of the model group.2.Western blot:The protein expression of nephrin,p-nephrin,Arp2/3 were markedly higher in the blank control group than in the model group (both P<0.01) and were lower in the model group than in the each Danshao group (both P<0.05).ConclusionDanshao Granular IH can effectively increase the products of nephrin,p-nephrin,arp2/3 and maintain podocytes signaling pathways and then reduce renal damages,improve the symptoms of HSPN in rats.
