Objective:Based on the abnormal bowel of dysfunctional constipation model, the effect difference induced by EA chang he point, mu point, he and mu points would be explored, and the change of the enteric nervous system inhibitory motor neurons before and after EA would be observed. The relationship between the effect difference evoked by EA at different acupoints regulating bowel function and enteric nervous system inhibitor motor neurons would be analyzed. It would provide reference for use of acupoints selected and combined reasonably to treat bowel diseases and symptoms in clinie.Methods:110 Sprague-Dawley male rats 4-week-old were randomly divided into normal control group 20 and model group 90. The model group rats were given by gavage with 0-4℃ ice water during the making the model period 14 days. The wet weight feces, dry weight feces, the first grain defecation time and small intestine propulsive function were observed and measured. On the 15th day, the models made successfully were randomly devided into four groups after the end of modeling. The groups included model group 12 rats and three EA groups (EA he point, EA mu point and EA he and mu point,6 rats in each group). The EA intensity was 1.5mA and the frequency was 2/15HZ. Each rat was treated by EA for 15min everyday in three EA groups. The wet weight feces, dry weight feces, the first grain defecation time and small intestine propulsive function were also calculated and analyzed. On the 22nd day, the rats colonic tissue sample were made and fixed from normal control group, model group and three EA groups. The expression NOS in both proximal and distal colon myenteric nerve plexus were detected by immunohistochemistry and the positive reaction analyzed with density mean.Results:(1) In modeling period, both of the wet weight feces and dry weight feces in model group decreased, the wet weight feces dropped significantly on day 2,8,12,14 compared with normal control group(p<0.05), and the dry weight feces was lower than normal control group(p<0.05). The ice water intragastric administration has little influence on the rats intestine function, but the first grain defecation time was lower than the normal control group(p<0.05).(2) In the EA treatment period, three EA groups could make the wet weight feces recover near normal level, and the effect of EA at he and mu points was superior to that in EA he point group(p<0.05). All three EA groups could make the dry weight feces recover near the normal level and also three EA groups could improve the first grain defecation time. EA at mu point could make the colon function recover near normal level, but could not make it return to normal level. The effect of EA at mu point to improve the colon function was weaker than EA at he point and EA he and mu points.(3) In the proxinmal colonic myenteric plexus, the nNOS in model group expressed significantly higher than that in normal control group(p<0.01). After EA intervention, all three EA groups can make abnormal nNOS high expression in model group constipation rats proxinmal colonic myenteric plexus decreased significantly(p<0.01), and EA at he point and EA at he and mu point could also make nNOS expression return to normal level except EA at mu point. The difference was statistically significantly between EA mu point group and EA he and mu group(p<0.01).(4) In the distal colon myenteric plexus, the expression nNOS in model group obviously increased than that in noamal control group(p<0.01). All three EA groups can make the abnormal nNOS high expression decreased compared with that in model group(p<0.01), and EA at he point could make nNOS return to normal level except EA at mu point(p<0.01). It shows that the effect of nNOS indistal colon myenteric plexus might play a more vital role in EA at he point and EA at he and mu points in regulating constipation rats bowel function.Conclusion:(1) Intragastric administration with ice water successfully duplicating the functional constipation model is verified again in the study. The feasibility and advantage of intragastric administration with ice water can enable the experimental period short and be simply carried out. Inhibitory motor neurons nNOS in both proximal and distal colon myenteric plexus abnormally increase result in the occurrence mechanism of functional constipation.(2) EA at he point, EA at mu point and EA at he and mu points can improve colonic function, and enable bowel function recover near normal level. It shows that, in constipation state, the effect generated by the combination of two jiyuan acupionts is better than that generated by single acupoint. The anxoaction generated by EA at he and mu acupoints regulating the constipation model rats colon is possibly resulted from the effect produced by the sacral parasympathetic nerve activated more easily. Therefore, it could enhance the effect on the combination of two jiyuan acupoints.(3) The effect of EA at mu acupoint regulates inhibitory motor neurons nNOS in both proximal and distal colon myenteric plexus is poor, and the effect of EA at he and mu acupoints is the best, The effects generatated by EA at he, mu or he and mu acupoints are different. The different changes of inhibitory motor neurons nNOS in both proximal and distal colon myenteric plexus possibly might be a basis for taking part in the effect difference between mu point and he and mu acupoints regulating bowel function..
