Objective:Through the observation of mTOR protein, SOD, MDA, NO and aortic morphology changes in spontaneously hypertensive rats, to explore the way of mTOR pathway in moxibustion anti hypertensive rats blood vessel damage stress tolerance mechanism, in order to provided the new mentality to reduce high blood pressure target organ damage.Methods:12 weeks of spontaneously hypertensive rats were randomly divided into two groups: wheat model control group and moxibustion group,10 cases in each group. Another 10 WKY ratsof 12 weeks as blank control group. Blood pressure of every rats were measured before intervention, and record. Each group was given he same breeding environment. Wheat model control group and blank control group without moxibustion therapy, while the moxibustion group got moxibustion with seed-sized moxa cone at "zusanli" and "quchi" at the same time of both sides, each acupoint has 5 cones,1 times a day for 28 days. After the last therapy,measured blood pressure of all groups again and recorded. Used the method of orbital venous plexus took the blood collection in all rats, to measured the SOD、MDA、NO content in serum、After collected all rats’ blood, killed all the rats.Took their hearts first, then took the thoracic aorta into 4% formaldehyde.Tested levels of the expression of mTOR protein in cardiac muscle tissue by immunohistochemical method. Used the HE staining and masson staining two ways to observe thoracic aorta morphological changes of all rats.Results:1.Blood pressure:before the experiment when all rats for 12 weeks, the blood pressure of wheat model control group is higher than the blank control group (P<0.01)[194.51±9.75mmHgvs122.09±7.61mmHg the blood pressure of moxibustion group is higher than the blank control group(P<0.01)[192.49±11.17mmHgvs122.09±7.61mmHg]. wheat model control group and moxibustion group have no difference (P> 0.05). Measured the Blood pressure after the experiment, the blood pressure of moxibustion group is lower than the wheat model control group(P<0.01)168.54±8.31mmHgvs241.90±7.34mmHg].2.SOD content in serum of every groups:Compared with blank control group, the SOD content wheat model control group.decreased (P<0.01)[95.95±6.53U/mlvs47.54±4.10U/ml]; Compared with blank control group,SOD content in moxibustion group decreased(P<0.01)[95.95±6.53U/mlvs76.63±6.48U/ml] Compared with wheat model control group,SOD content in moxibustion group increased(P<0.01)[47.54±4.10U/mlvs76.63±6.48U/ml]3.MDA content in serum of every groups:Compared with blank control group, the MDA contenwheat model control group increased(P<0.01)[0.50±0.14nmol/mvs2.01±0.15nmol/m]; Compared with blank control group,MDA content in moxibustion group increased(P<0.01)[0.50±0.14nmol/mvs0.83±0.10nmol/ml]Compared with wheat model control group,MDA content in moxibustion group decreased(P<0.01)[2.01±0.15nmol/mlvs0.83± 0.10nmol/ml]。4.NO content in serum of every groups:Compared with blank control group, the NO content of wheat model control group decreased(P<0.01)[77.76±6.08umol/Lvs47.30± 7.05umol/L]; Compared with blank control group,NO content in moxibustion group decreased(P<0.01)[77.76±6.08umol/Lvs68.80±6.57umol/L]Compared with wheat model control group,NO content in moxibustion group increased(P<0.01)[47.30±7.05umol/Lvs68.80± 6.57umol/L]。5.The result of mTOR expression by imunohistochemical method:Compared with the blank control group,the mTOR protein expression of wheat model control group rats decreased (P<0.01). Compared with the blank control group, the mTOR protein expression of moxibustion group decreased (P<0.01)。Compared with the wheat model control group, the mTOR protein expression of moxibustion group increased(P<0.01)6.The result of aortic morphology changes by HE:In the blank control group,we could not see the obvious change of ortic morphology.we could see smooth intima and media thickness from inside to outside.At the same time,layer is clear and vascular smooth muscle cells is in normal form.In the wheat model control group:we could see that media became thick and more vascular smooth muscle cells appeared with arranged in rows. Shape and arrangement of the endothelial cells were irregular, suggested there has been a rat aortic vascular injury.In the moxibustion group, there is not obvious aortic intima hyperplasia, just some lining phenomenon happened. The membrane elastic fiber is irregular, part of them break, media thickness,and smooth muscle cells were in normal form.7.The result of aortic morphology changes by masson:In the blank control group,we could hardly see vascular collagen fiber.A large number of collagen fibers in wheat model control group.And we could see a few vascular collagen fibe in moxibustion group.Rate of fibrosis in vessel:Compared with blank control group, the rate of fibrosis of wheat model control group increased(P<0.01)[7.64±3.21%vs35.87±10.80%] Compared with blank control group,the rate of fibrosis in moxibustion group increased(P<0.01)[7.64±3.21%vs14.18±4.84%]Compared with wheat model control group,MDA content in moxibustion group decreased(P<0.01) [7.64±3.21%vs14.18±4.84%].Conclusion Moxibustion can change the level of oxidative stress and reduce the blood vessel damage by mTOR pathway.
