Background:colorectal carcinoma (CRC) is defined as one kind of malignant lesions occur in colorectal mucous epithelium because of all kinds of carcinogenic factors including environmental and genetic factors. The bad prognosis and high mortality of CRC is unfavorable. CRC is also one of the most common malignant tumor in China. CRC, including colon and rectum cancer, is the third-most malignant tumor. Patient with CRC is asymptomatic or their symptom is too unconspicuous to notice that they just feel a little uncomfortable including dyspepsia and defecate occult blood etc. As CRC develops, the symptoms becoming more and more severe and distinct including changes of diachoresis habit、 stomachache、hematochezia、abdominal mass、intestinal obstruction and so on。 Sometimes patients also show anemia fever and emaciation.According to the epidemiology statistics, the orbidity of CRC increases 4.2% annually. So far, the morbidity and mortality rate of CRC ranks the fifth of carcinomas in the world. When it comes to the pathogenetic and initiated mechanism, it stays ambiguous.Purpose:this research is to discuss the mobility of rats with yin and yang syndrome along with the experiment, and the expression of polarity proteins of Lgl、 vangl and podocalyxin in colorectum. Data would be used to analyse the relation among the three proteins and to understand the relation between the mobility of CRC and the three proteins. Finally, this research is to reveal the potential effect and clinical significance of Lgl、vangl and podocalyxin to CRC.Materials and Methods:156 rats were randomly divided into five groups:model group with yin syndrome, model group with yang syndrome, treatment group with yin syndrome, treatment group with yang syndrome and blank group, while the former four groups are consist of 36 rats respectively, and blank group is consist of 12 rats. Each group is half male and half female. The two groups with yang syndrome were administrated by gavage with ethanol solution for 3 weeks and fed with extra capsaicin and ethanol solution for another 2 weeks. The two groups with yin syndrome were administrated by gavage with cold water for 5 weeks and extra iced water bath in the last 2 weeks. After the yin and yang syndromes of animal models were built, rats were subcutaneously injected with DMH (1, 2-DIMETHYLHYDRAZINE DIHYDROCHLORIDE) once a week for 12 weeks. During the same time, treatment groups with yang syndrome and yin syndrome were administrated by gavage with Zuojinwan and Fanzuojinwan once a day respectively for 24 weeks, while the animal models with yang and yin syndrome were still being built, but yang syndrome group was built by means of administration by gavage with ethanol solution and yin syndrome group was built by administration by gavage with cold water. In the fifth week,36 rats were dissected to see the condition of yin and yang animal model. After injected with DMH for 12 weeks,24 rats were dissected every 3 weeks to observe the morbidity of CRC. Lgl、vangl and podocalyxin are detected in normal and cancerous colorectum to comPare the difference of expression.Result:After modeling the yin and yang syndrome, rats show obvious yin and yang signs and symptoms which means the success in modeling. Morbidity of CRC of treatment groups is lower than model groups which means that traditional Chinese medicine is beneficial for CRC treatment. Immunohistochemical detection and pcr detection reveal that the expression of polarity protein of Lgl、vangl and podocalyxin is different in normal colorectum and colorectal carcinoma. The expression of Vangl and podocalyxin in CRC is higher than normal tissue while the expression of Lgl in CRC is lower than normal tissue. It means that the three polarity protein is closely associated with the genesis and development of CRC.Conclusion:1、The animal model with yin syndrome can be built by means of administration by gavage with cold water and extra iced water bath.2、The animal model with yang syndrome can be built by means of administration by gavage with capsaicin and ethanol solution3、The expression of polarity protein of vangl、Lgl and podocalyxin in normal colorectum and CRC is different, which means that these three polarity proteins are closely associated with the genesis of CRC.
