Objective:Investigate the pharmacologic action of astragale powder to leptin defiency ob/ob mice insulin resistance,confirm the hypothesis about astragale powder mediate "leptin-insulin" balance and ameliorate insulin sensitivity through adipoinsular axis.Methods:Use Ob gene mutation leading to leptin defiency ob/ob type 2 diabetic mice as model,littermates wild type C57BL/6J mice as control,investigate the influence of astragale powder to ob/ob mice food intake,body weight,plasma glucose,plasma lipid,glucose tolerance,insulin tolerance and plasma leptin,and treat with exogenous recombinant murine after 14 weeks,observe the influence of astragale powder and leptin to mice insulin sensitivity and glucolipid metabolism of organs,and try to analyse possible mechanism.Results:Astragale powder failure to decrease ob/ob mice glucose and ameliorate insulin resistance;after leptin treatment,ob/ob mice food intake,body weight decreased significantly,insulin sensitivity obviously enhanced,plasma insulin level reduced significantly,hepatic and adipose tissue lipid content decreased significantly,hepatic glycogen content increased,the mRNA expression of JAK2,STAT3,IRS2,PI3K,Akt in liver enhanced significantly,the mRNA expression of FAS,LPL,MCP1 in subcutaneous fat decreased significantly.Conclusion:According to previous research of astragale powder attenuates STZ+high-fat-diet(HFD) induced type 2 diabetic B6 mice body weight gain,hepatic and adipose tissue lipid content,decreased plasma leptin and insulin level,attenuates insulin resistance,and astragale powder failure to improve the leptin defiency ob/ob mice and leptin almost defiency AP2-SREBP1C mice insulin sensitivity, confirm astragale powder ameliorate insulin resistance through mediate "leptin-insulin" balance,through enhance leptin snsitivity,then improve insulin sensitivity,and decrease body weight,promote lypolysis, attenuates peripheral tissues steatosis,and combine the results of astragale powder decreased diabeitc B6 mice PEPCK expression in liver,enhanced ob/ob mice JAK2,STAT3,IRS2,PI3K,Akt mRNA expression in liver and inhibit FAS,LPL,MCP1 mRNA expression in subcutaneous fat,we postulate astragale powder mediate endocrine ability of adipose tissue through adipoinsular axis,decrease leptin secretion and improveleptin’s sensitivity,decrease peripheral tissues lipogenesis,enhanced lypolysis, attenuates steatosis,and improve hepatic insulin sensitivity through mediate "leptin-insulin" signal pathway,treat the type 2 diabetes.
