| Objective:Dendritic cells(DCs) are the most powerful antigen presenting cells as now known, which play key roles in the initiation and amplifying of innate and adaptive immune response. Different stages of differentiation DCs different athletic ability, which could lead to an immune response results. Therefore,Discussion of dendritic cells in different stages of differentiation cytoskeleton binding proteins influence. It is significant for further understanding the DCs exercise capacity as well as improving the clinical efficiency of DCs-based cancer immune therapy.Methods: Monocytes from perphrial blood were induced grown into im DCs by rhIL-4 and rhGM-CSF for 5 days as well as mature dendritic cells(mDCs) were induced by TNF-α(tumor necrosis factor-α), GM-CSF and IL-4 for 3 days in vitro.Extract the total protein and total RNA from dendritic cells at different differentiating stages, of which the m RNA and protein levels invovled in cytoskeleton were detected by Real-time PCR and protein chip. Results:At the processes of monocytes differentiation into mDCs via imDCs, the expression levels at gene and protein of some monomer-stabilizing proteins, capping proteins, cross-linking proteins,actin-depolymerizing proteins, nucleating proteins, membrane-bound proteins,microtubule-associated proteins and motor proteins were up-regulated or down-regulated in different degrees.The results of PCR are consistent with those of protein chip. Conclusion:In conclusion, the study found that some DCs skeleton binding proteins in DCs at different differentiation stages displayed various expression levels, which were involved in cell skeleton remodeling, migration,adhesion, phagocytosis and deformation. The structure and function of DCs by the synergistic effect of these proteins and tightly regulated, but the specific mechanismneeds further study, it’s significant for further understanding the immune function of DCs. |
