Objective:To prove the protective effects of S1 P on male mouse reproductive cells by toxicity damage and study the mechanism of S1 P. Methods:Forty healthy male Kunming mice were randomly divided into 5 groups(the normal group, the low,middle and high dose group of S1P( 0.5,1.0,2.0μg/10g),the cyclophosphamide positive control group).The normal group were given normal saline(0.1ml/10g) and the positive control group were given cyclophosphamide(40mg/kg).Mice in each group were given intraperitoneal injection for 5 days. The mice were killed at the thirtieth day after the first treatment. We collected serum,sperm suspension,epididymis,testis of the mice respectively. Then we counted the sperm number, activity ration and deformity.We determined the activity of LDH by colorimetric method in each group of mice.To observe pathologic changes of testis by pathological section. To detect Bcl-2, BAX and Caspase-3expression by immunehistochemical staining and determine cell DNA damage by comet assay. Results : Compared with the positive control group, there existed significant differences in the sperm number,deformity,the activity of LDH,the pathologic changes,Bcl-2,BAX,Caspase-3 expression and cell DNA damage of each experimental S1 P group(P<0.05). Conclusion:There were some protective effects of S1 P on male mouse reproductive cells by toxicity damage. Maybe the mechanism is S1 P can inhibit apoptosis protein to reduce the DNA damage by toxicity.
