Objective:Dry eye is the most common ocular surface disease, it has become a worldwide health problem.There are many mechanism of action of dry eye.At present, most scholars believe that inflammation is a key problem in the occurrence of dry eye.This research aims to make the animal model of dry eye induced with inflammatory factors and observe the influence"Xiao Sheng San" making to dry eye modelassociated inflammation factors.We hope it could provide new choose of the formulas for treatment of related diseases and the basis for later development of new dosage forms.Methods:1.The design of dry eye animal model:select 5 weeks of healthy female C57BL/6 mice,Subcutaneous injection of scopolamine 0.5mg(The concentration of 5 mg/ml),four times a day,and placed in a continuous air flow environment,Humidity<35%,10days consecutively.2.Observe the intervention effect of Xiao Sheng San preparation on dry eye model induced scopolamine:the research object is dry eye model in the C57BL/6 mice induced by scopolamine,we observe tear secretion,tear break-up time and corneal fluorescein staining. Observe different concentrations of "Xiaosheng San " effects of the cornea conjunctival tissues morphology and inflammatory factors.3.Discussion on mechanism of action of "Xiaosheng San" in the control of inflammation mechanism:After modelling,the eyeballs of the rats were taken out, after the traditional paraffin section, the expression of inflammatory cytokines in the cornea conjunctival tissue were observed using Immunohistochemistry.Results:l.dry eye model induced scopolamine for 7 days、14 days and 21 days grouphave obvious differences with the normal group in tear secretion,tear break-up time and corneal fluorescein(P<0.05), but no difference in three model groups,it prompts that the animal model of mice is long-term stability.2.Low concentration、medium concentration and high concentration "Xiaosheng San"have statistical differences with model group in above test items at 7 days、14 days or 21 days.3.From the results of HE staining and immunohistochemistry, The cornea conjunctival tissue organization form of the model group changed obviously with the normal group,corneal epithelial cell shape is irregular, levels not neat, irregular stromal layer cell edema,IL-1 inflammatory factors increased significantly,low, middle and high "Xiaosheng San" groups improved the morphology of the cornea and reduce inflammation factors.Conclusion:1."Xiaosheng San"dry eye has certain therapeutic effect with model induced scopolamine.the middle concentration "Xiaosheng San"groups improved the most obvious, followed by high concentration group.2.The mechanism of action of "Xiaosheng San" may related to the control of ocular inflammation.3.The research results show that"Xiaosheng San" has a protective effect with the cornea conjunctival tissue, but need to further research.
