| Aging population is the development trend of the world today. The structure of the tissues and organs of the body have changed with the age increasing while the function have declined,and the health status is gradually decreasing.In which the occurrence of degenerative disease in the blood vessels with aging is called vascular aging.Studies have shown that vascular aging is a risk factor for older people to suffer from cardiovascular disease. Therefore, it is significant to study the correlation of vascular aging for the prevention and treatment of cardiovascular diseases.The cytoskeleton is a dynamic protein network system in eukaryotic cells, which participates in many cellular functions.Cytoskeletal integrity and stability are essential for maintaining the morphology of vascular cells and cardiac muscle, and the changes of the structure will affect the function of vessel and myocardium. This experiment, which aims to further explore the cytoskeleton changes of vessel and myocardium during aging process, and the intervention effect of Chinese medicines of tonifying Qi and activating blood circulation by having established the natural aging rats model and carried on related research, will provide a new basis for anti-aging of traditional Chinese medicineThis study is divided into two parts:literature review and experimental study.1. Literature review:The paper,which summarizes the cytoskeleton changes of during vascular aging, and the research to delay vascular aging and aging by the method of tonifying Qi and activating blood, indicates that the change of cytoskeleton can affect the structure and function of vessels,and induce the corresponding vascular diseases.2. Experimental study:Objective:To observe the carotid artery and myocardium morphological and cytoskeleton changes during the senescence, further explore the mechanism of vascular aging and give evidence to the interaction effect of Chinese traditional medicine(extracts from Radix Ginseng, Radix Notoginseng and Rhizoma Chanxiong).Methods:The model aging rats were randomly divided into 5 groups, respectively for aging model group, Chinese medicine low-dose, medium-dose, high-dose group and resveratrol group,and another rats as young control group. The high,middle and low daily dose of Chinese medicine are followed by 2.32g/kg/d〠1.16g/kg/dã€0.58g/kg/d and resveratrol group dose of 0.05g/kg/d, aging model group and young control group given the appropriate volume of distilled water. Administered orally,1 times a day.After 3-month intervention,the following study were oberved observation of aging rats behavior change through open field test, detection thymus index and spleen index,detection the content of rat serum ROS, MMP-2, MMP-9, aging and morphological changes of carotid and myocardium by P-gal and HE,observation the distribution of carotid collagen fiber by masson, detection the content of a-tubulin, p-tubulin,F-actin and HSP 27 of carotid and myocardium by immunofluorescence,immunohistochemistry and Western blot.Results:Compared with the young group,aging rats is dull and depression with the yellow and lacklustre hair.cross grid total number were reduced, head diameter distance decreased, the total number of cross cell(P<0.01) and the total action distance (P<0.05) decreased,thymus index and spleen index decreased(P<0.01).Carotid and myocardium β-gal positive expression area increased.Aging carotid artery changes including wall thickening, non smooth intima, disorder of smooth muscle cells, elastic membrane distortion and obvious hyperplasia of the connective tissue of the outer membrane.Vascular collagen area ratio increased (P<0.01).Aging myocardium thickening, fibers disorder, and even the emergence of myocardial fibers dissolved fracture phenomenon. The serum levels of ROS, MMP-2, MMP-9 increased (P<0.05); the mean fluorescence intensity and average optical density values of a-tubulin, P-tubulin,and F-actin in carotid and myocardium increased (P<0.01); protein relative expression values of a-tubulin, β-tubulin,and F-actin increased (P<0.01), HSP27 protein relative expression value decreased (P<0.01)in aging model group. Compared with the aging model group,the general state of aging rats using chinese medicine and resveratrol improved, the cross lattice of aging rats and the total number of the total act distance increased (P<0.05),The coefficient of thymus coefficient in middle, high dose of Chinese medicine group and resveratrol group increased in rats (P<0.05), the coefficient of spleen in high dose of Chinese medicine group and resveratrol group increased(P<0.01).In all Chinese medicine groups and resveratrol group, β-gal positive expression area of Carotid and cardiac decreased, aging carotid and cardiac remodeling improved, collagen fiber in vessel wall area ratio decreased (P<0.05);the serum ROS, MMP-2 content decreased (P<0.05);the serum MMP-9 decreased (P<0.05)in medium and high dose Chinese medicine group and resveratrol group.The mean fluorescence intensity and average optical density values of a-tubulin, β-tubulin,and F-actin in carotid and myocardium decreased (P<0.01); protein relative expression values of a-tubulin,β-tubulin,and F-actin decreased (P<0.01), HSP27 protein relative expression value increased(P<0.01).Conclusions:1.With the increase of age, the overall trend of behaviors, immune function,oxidative stress and extracellular matrix changed,including decline in the total number of acrossinggrid, the total behavior distance of rats, thymus and spleen coefficient, and ROS,MMP-2, MMP-9 increase in serum.2. With the increase of age, the carotid artery and myocardium of rat changed, including the performance of vascular wall thickening, myocardial hypertrophy and other vascular, myocardial remodeling phenomenon, in addition, the area of collagen distribution increase.3.With the increase of age,the cytoskeleton of the rat carotid artery and myocardium changes, the content of a-tubulin, β-tubulin,and F-actin increased, HSP27 content decreased.4.The extracts from Radix Ginseng, Radix Notoginseng and Rhizoma Chanxiong and resveratrol can improve aging rats behavioral ability and immune function, reduce oxidative stress and matrix metalloproteinase levels, improve vascular and myocardial remodeling.Chinese medicines of tonifying qiand activating blood and resveratrol may through the HSP27 down F-actin content, reverse the change of cytoskeleton components, and then affect the vascular and cardiac function of the skeleton for delaying aging. |
