Objective Human parvovirus B19 (B19V), Cytomegalovirus (CMV) and Herpes simplex virus (HSV) can be spread through blood transfusion, to establish the validity of quantitative real time PCR(Q-PCR) methods based on SYBR Green for detecting the prevalence of these viruses, and measuring the viral load in healty blood donors. At the same time, to investigate the seroprevalence of B19V, CMV and HSV in healthy Chinese blood donors in five blood centers.Methods Building a quantitative real-time PCR system to detect DNA in plasma from five Chinese blood center, including Chongqing, Luoyang, Urumqi, Mianyang and Liuzhou, and measuring the viral load of positive samples. In addition,292 plasma were randomly selected from five blood centers to detect for IgG and IgM infections using ELISA method.Results The sensitivity of the established Q-PCR system is high with B19V, CMV and HSV. The limit of detection (LOD) for B19V is 5 cp/mL and for CMV and HSV was 50 cp/mL In this group of 3794 blood donors population,21 B19V DNA positive samples,6 CMV DNA positive samples and 43 HSV DNA positive samples were detected, accounted for 0.55% (21/3794),0.16%(6/3794) and 1.13%(43/3794), respectively. In 292 specimens by ELISA, IgG antibody positive rate for B19V, CMV and HSV was 23.6%(69/292),37.3%(109/292) and 16.44%(48/292), respectively. IgM antibody positive rate was 1.71%(5/292),8.9% (26/292) and 4.45%(13/292), respectively. IgG and IgM antibody positive rate was 0.68% (2/292),1.71%(5/292) and 2.40%(7/292).Conclusion There is a certain prevalence of B19V, CMV and HSV in our country, because of the virus in immunocompetent individuals are asymptomatic infection, but in immunocompromised patients, organ transplant patients, newborn baby and pregnant woman exist for a long time, it is recommended that these special populations before blood transfusion or organ transplantation should be be clear whether there is a virus infection in receptor and acceptor, and adjust the clinical treatment.
