In the viral vaccine research and development work, human diploid cell matrix prepared as viral antigen is the first choice of the entire vaccine development and production quality control. The correlation analysis of genetic stability and virus infection biology reaction characteristics of human diploid cell as the vaccine preparation matrix provide more abundant information for research and development of virus vaccine, including vaccine safety studies.From the point of view of cell biology, cell specific enzymatic reaction characteristics are believed to reflect genetic characteristics and biological properties. Existing data indicates the facts. When the cells are faced with virus infection, the natural immune response to the biological response is in fact related with its own biological traits. The interferon response produced by cells has a significant mass index in the vaccine preparation process technology, and the index has a direct correlation with the biological activity state in cell proliferation. Therefore, the analysis of cell enzyme activity and the intensity of interferon response in virus infection have important significances to cell research prepared for vaccines. So this paper uses a variety of cell lines, multi-generation cells, detecting of the enzyme activity of intracellular lactate dehydrogenase, superoxide dismutase, succinate dehydrogenase by biochemical methods to identify the stability of cells. Experimental results show that the intracellular enzyme activity has some changes with cell passages. when human embryonic lung diploid cells (KMB17 cells) and new human embryonic lung diploid cells (No cells) in the cell passages are over the 30th generation, the activity of lactate dehydrogenase and succinate dehydrogenase increases, the activity of superoxide dismutase decreases. The results indicate that the ability of cell proliferation and metabolism drops and the cells have poor stability when the two kinds of cells in the cell passages are over the 30th generation. So they are not conducive for the development and production of vaccines when the two kinds of cells in the cell passages are over the 30th generation. With increased cell passages, Vero cells and Hela cells have no significant changes in enzyme activity, it suggests that the ability of cell proliferation and metabolism of these two kinds of cells remain stable during the experimental cell passages.This paper focuses on the mutual connection between the various enzyme activity index and the cell interferon response by the RNA and DNA viruses infection, and these links lead to virus titer changes. When HSV-1 virus as a complete proliferation in the nucleus and EV71 virus as a complete proliferation in the cytoplasm infect the human embryo lung diploid cell (KMB17, No), the results are that the relative expression of IFN-a has a great relationship with cell’s enzyme activity and cell growth state. When human embryo lung diploid cell are cultured for 3 days,cell enzyme activity is on the rise and the growth state of the cells is better. when the cells are cultured for 5 days, cell enzyme activity is in a steady state and the growth state of the cells is maintained relatively stable. Both viruses infected the cells which are cultured for 3 days cause higher relative expression of IFN-a than those infected the cells which are cultured for 5 days. The viral titer gradually increase during infection, but its proliferation gradually decreases with the increase of the secretion of interferon. The fact suggests that the limit of accommodation of human diploid cells in the virus proliferation may be related with interferon concentration.When both viruses infect the Hela cells which are cultured for 3 days and 5days, they cause the relative expression of IFN-a have little relationship with cell enzyme activity. At the same time, relative expression of IFN-a are lower than other cells. This suggests that the cell is a non-restrictive feature as the tumor cell. On the contrary, both viruses infected the Vero cells which are cultured for 3 days cause higher relative expression of IFN-a than those infected the cells which are cultured for 5 days. And virus titer are consistent with the above changes. The feature is different from human diploid cells, which seems to prompt the biological characteristics of Vero cells.The work is that studying the relationship between the relative expression changes of IFN-a and cellular enzyme activity、cells growth state, although this requires further confirmation and analysis, but the associated data provides the experimental basis for vaccine research and preparation.
