Cancer theranostics can simultaneously assess the disease state of individual patients, and according to the diagnostic results can also tailor the therapeutic treatment, so it hopes great significance for personalized medicine. Ideal cancer theranostics should possess the imaging and therapy modalities, and it can generate prominent imaging and high therapeutic efficacy in the disease site, but has small side effects for normal tissues.The rapid development of nanotechnology has brought new opportunities for cancer theranostics. Quantum dots(QDs) because of their unique optical and electronic properties have been widely used in biological imaging, while the QDs can be used as drug carriers, and the photosensitizers can be conjugated onto them, then the QDs will transfer the energy to the photosensitizers for photodynamic therapy to achieve imaging and therapy modalities.In this paper, we synthesis the water-soluble QDs, and construct a two dimensional molecular beacon(TMB) probe with a photosensitizer chlorin e6(Ce6), a QD and a dark quencher BHQ3. The TMB can be specifictly activitated by the tumor mRNA of cancer cell lines and xenografted tumor models, then the imaging and therapy modality are generated by the FRET between the QD and the dark quencher BHQ3, the FRET between the QD and the photosensitizer Ce6. Compare to the traditional photosensitizer Ce6, the TMB probe has appreciable imaging and therapeutic efficacy, which realize the cancer theranostics.
