Objective To make a further define what Th1/Th2/Th17/Treg role in the pathogenesis of SSc,we plant to detect the levels of Thl/Th2/Th17/Treg cells related cytokines IFN-γ,IL-4,IL-17A and IL-10 in the serum of SSc patients, and analyze the correlation with cytokines and MRSS scores,HRCT scores,disease activity scores or disease severity scores.Methods By collecting clinic data and blood specimens from 31 cases of patients with SSc, and using Enzyme linked immunosorbent test (ELISA) to detect levels of IFN-γ,IL-4,IL-17A and IL-10 in the serum and compare,to analyze the correlation with cytokines and MRSS scores,HRCT scores,disease activity scores or disease severity scores.Results1.The level of IL-17A in the serum of the SSc patients show higher than the healthy controls (22.53±17.49 VS 14.61±5.96 ng/ml,P<0.05), the level of IFN-γ,IL-10 show lower than the healthy controls (10.95±5.08 VS 15.72±6.94,1.81±1.23 VS 3.14±2.54ng/ml,P<0.05) however IL-4 show no statistical difference with the healthy controls (4.16±1.90 VS 4.49±2.67 pg/ml,P>0.05).2.The level of IFN-y in the serum of the SSc patients are negative correlated with MRSS scores (r=-0.369,P=0.045),and show no significant correlation with HRCT scores,disease activity scores or disease severity scores(P>0.05). IL-17A are positively correlated with disease activity scores (r=0.523,P=0.003),show no significant correlation with HRCT scores,MRSS scores or disease severity scores(P>0.05).IL-4,IL-10 were correlated with MRSS scores,HRCT scores, disease severity scores or disease activity scores(P> 0.05).3.IL-17A in the healthy controls, the inactive group were lower in the activite group (P<0.05);IFN-y in the inactive group and the activite group were lower than the healthy controls (P<0.05),show no difference between activite group and the inactive group (P>0.05);IL-10 in the inactive group is reduced to the activite group (P<0.05), show no difference between activity group and the healthy controls (P>0.05);IL-4 there is no obvious difference between groups (P>0.05).4.IL-17A/IL-10 in the serum of the SSc patients show higher than the healthy controls (Z=-3.008, P=0.003),however, IL-4/IFN-γ show higher with the healthy controls,but no statistical difference (Z=-1.698, P=0.090).Conclusion1.The level of IL-17A in the serum of the SSc patients is increases, and the activite group was higher in the healthy controls and the inactive group, and was positively correlated with disease activity scores, which indicate the level of IL-17A in serum can be used as index of assessment of disease activity.2.The level of IFN-y in the serum of the SSc patients show lower than the healthy controls and negative correlated with MRSS scores, the decline of IFN-γ maybe correlated with the degree of SSc patients’ skin fibrosis.3.The level of IL-17A in the serum of the SSc patients is increases,and IL-10 declining, IL-17A/IL-10 in the serum of the SSc patients show higher than the healthy controls, which indicate the Th17/Treg cell imbalance may exists in the SSc patients.
