Objective To explore the protective effects of recombinant human erythropo ietin on hyperoxic cerebral injury in neonatal rats.Methods Ninety neonatal wistar rats were randomly divided into three grou ps:air-exposed control group (Al group), hyperoxia-exposed and placebo grou p (80μ2% oxygen) (A2 group), hyperoxia-exposed and recombinant human e rythropoietin (rEPO) treated group (A3 group), n=30/group.The assessment of hippocampus tissue changes by hematoxylin and eosin staining (n=10/group, eve ry time point).Caspase-1, Caspase-3, Interleukin-18 and B cell lymphoma-2 lev els were measured by western blot.Result 1. Hippocampus tissue of group A2 had different levels on swelling andapoptosis of cell with inflannatory reaction by ematoxylin and eosin stainin g under the light microscope and the extent of injury were more and more seri ous with exposure of hyperoxia time extension, the degrees of brain injury of e very time point on group A3 were more relieve than group A2.2.Western blotting showed that the protein expression of caspase-1, caspase-3 and interleukin-18 on group A2, A3 were.up-regulation compared with group Al with extending hyperoxia exposure time, and theirs gray levels were outstan ding rise.The protein expression of group A3 was decrease compared with group A2.Western blotting showed that the protein expression of B cell lymphoma-2 on group A2,A3 were down-regulation compared with group Al with extending hyperoxia exposure time, and theirs gray levels were outstanding decline.The sig nificant difference of the protein expression of caspase-1, caspase-3, interleukin-18 and B cell lymphoma-2 between group A2 and group A3 on the seventh day and fourteenth day in statistics (P<0’.05)Conclusion Our result suggest that rEPO generates its protective effect agai nst the apoptosis factor.The mechanism is down regulation protein expression of caspase-3 and is rise regulation protein expression of B cell lymphoma-2 relate d to endoplasmic reticulum stress.And it is down regulation protein expression o f caspase-3 related to inflammatory reaction and inflammatory factor interleukin-18...
