Objective:To detect the changes of inflammatory factors in diabetic rats serum IFNg, MCP-1, TNF-a, IL-1α, IL-1β,IL-2, IL-4, IL-6, IL-10 concentration.methods:There are 24 Male Sprague Dawley (SD) rats aged 8 weeks, randomly selected 8 rats as normal control group (control group), and the remaining 16 rats were fasted for 12 hours and then they were induced by a single intraperitoneal injection of streptozotocin(STZ) at a dose of 55mg/kg. The rats with random blood glucose above 16.7mmol/L were regarded as successful diabetic rats model and were randomly divided into two group: DM+NRG-1β group(n=8) and diabetic control group(n=8). The DM+NRG-1β group were treated with injection of rhNeuregulin-1β at a dose of 10μg/kg.d for consecutive 10 days and the diabetic group with simultaneously administrating with O.lmmol/L citrate buffer (PH:3.0). The serum concentrations of IFNg, MCP-1, TNF-a, IL-la, IL-1β, IL-2, IL-4, IL-6, IL-10 in each rat were measured using sandwich chip technology (RayBiotech chip). Analyzing and processing data with Analysis of variance and SPSS 19.0.Result:1.The DM group compared the control group, significant difference in the serum concentration of IL-1α were detected between the two groups of rats[(6955.36±1234.52pg/ml)vs(2324.69±1667.45pg/ml), P=0.010] no significant difference in the serum concentration of IFNg were detected between the two groups of rats[(11.20+9.55pg/ml) vs (12.10+14.94pg/ml), P>0.05]; no significant difference in the serum concentration of MCP-1 were detected between the two groups of rats[(2234.02+1859.41pg/ml vs 2971.85+ 2364.50pg/ml), P>0.05]; no significant difference in the serum concentration of TNF-α were detected between the two groups of rats[(227.50±124.17pg/ml) vs (210.37±70.00pg/ml), P>0.05]; no significant difference in the serum concentration of IL-1β were detected between the two groups of rats[(1675.53±2155.99pg/ml) vs (1406.32±2532.02pg/ml),P>0.05];no significant difference in the serum concentration of IL-2 were detected between the two groups of rats[(10992.14±9497.73pg/ml) vs (14830.30±10814.49pg/ml), P>0.05]; no significant difference in the serum concentration of IL-4 were detected between the two groups of rats[(8.68±8.43pg/ml)vs(9.28±14.37pg/ml), P>0.05]; no significant difference in the serum concentration of IL-6 were detected between the two groups of rats[(518.00±323.19pg/ml) vs (689.96±345.76pg/ml), P>0.05]; no significant difference in the serum concentration of IL-10 were detected between the two groups of rats[(5874.33±4926.23pg/ml) vs (7466.91±5026.55pg/ml), P>0.05].2.The DM+NRG-1β group compared the DM group, significant difference in the serum concentration of IL-la were detected between the two groups of rats[(1888.51±1409.90pg/ml) vs (6955.36±1234.52pg/ml), P=0.010]; no significant difference in the serum concentration of IFNg were detected between the two groups of rats[(6.00±4.37pg/ml) vs (11.20±9.55pg/ml), P>0.05]; no significant difference in the serum concentration of MCP-1 were detected between the two groups of rats [(1127.07±565.51pg/ml) vs (2234.02±1859.41pg/ml), P>0.05]; no significant difference in the serum concentration of TNF-a were detected between the two groups of rats[(144.75±124.57pg/ml) vs (227.50±124.17pg/ml), P>0.05]; no significant difference in the serum concentration of IL-1β were detected between the two groups of rats[(2410.94±5120.51pg/ml) vs (1675.53±2155.99pg/ml), P>0.05]; no significant difference in the serum concentration of IL-2 were detected between the two groups of rats[(7835.04±3715.13pg/ml) vs (10992.14±9497.73pg/ml), P>0.05]; no significant difference in the serum concentration of IL-4 were detected between the two groups of rats[(6.47±5.40pg/ml) vs (8.68±8.43pg/ml), P>0.05]; no significant difference in the serum concentration of IL-6 were detected between the two groups of rats[(406.96±295.92pg/ml) vs (518.00±323.19pg/ml), P>0.05]; no significant difference in the serum concentration of IL-10 were detected between the two groups of rats[(3552.37±1097.78pg/ml) vs (5874.33±4926.23pg/ml),p>0.05].Conclusion:1. Serum concentrations of IL-1α in diabetic rats was significantly increased, while serum concentration of IL-la diabetic rats was significantly decreased after NRG-1 treatment; 2. Serum concentrations of IFNg, MCP-1, TNF-a and IL-1β, IL-2, IL-4, IL-6, IL-10 there was no significant difference, but in diabetic rats after NRG-1 beta treatment, serum concentration of IFNg, MCP-1, TNF alpha have had the trend of low, serum concentration of IL-2 have the tendency to increase.
