Objective:Liver fibrosis is a variety of pathogenic factors effect on the liver,leading to excessive deposition of extracellular matrix,intrahepatic fiber connective tissue hyperplasia, eventually leading to liver function and abnormal changes of structure of the disease process.The study found that,Hepatic stellate cell is the effect in the process of liver fibrosis, is also a target cells in the treatment of liver fibrosis.Transforming growth factor beta 1 and Smads signal pathway is Chinese traditional medicine the main signaling pathways anti liver fibrosis.Wet and heat, poison, blood stasis, phlegm, is the basic pathological factors in the process of the occurrence of liver fibrosis, "Wet heat","Blood stasis","Virtual"coexistence is the basic pathogenesis characteristics of liver fibrosis.Modified Yinshao pulvis is based on the principle of heat clearing damp invigorate the circulation of the centralizer stand by side,Collection of exorcism and centralizer is an organic whole.Previous studies found that modified Yinshao pulvis can inhibit duck hepatitis b virus replication, delay the process of liver fibrosis.Therefore explore modified Yinshao pulvis effect mechanism of TGF-beta 1/ Smads pathway and its influence on hepatic stellate cell proliferation,will provide theoretical basis for clinical treatment of liver fibrosis and various treatments.Aim:This experiment aims to observe modified Yinshao pulvis serum intervened hepatic stellate cell proliferation and TGF-betal,Smad3mRNA and protein expression.Further explore the mechanism of action on anti liver fibrosis which is based on the method of heat and damp clearing and invigorate the circulation of blood,Provide theoretical basis for clinical treatment of liver fibrosis and various treatments.Methods:adult male Wistar rats were given hexanone theobromine lavage Chinese medicine and physiological saline, adaptive feeding after a week, from the abdominal aorta in pursuance, preparation of drug serum. Serum set to six groups,Chinese medicine high dose (Yinshao pulvis bulk drug serum high dose intervention group),traditional Chinese medicine middle dose (Yinshao pulvis bulk drug serum middle dose intervention group), traditional Chinese medicine (TCM) low dose group (Yinshao pulvis bulk drug serum low doses of intervention group) and the negative control group (normal saline serum intervention group)and positive control group (hexanone theobromine serum intervention group) and blank group, serum drug intervention HSC-16, with a determined by MTT method to detect cell proliferation and flow cytometry instrument for testing, and PCR, Western blot detection Smad3, TGF-beta 1 mRNA and protein expression.Results:Modified Yinshao pulvis dispersion effectively inhibit rat hepatic stellate cells proliferation and TGF-beta 1、Smad3 mRNA and protein expression is restrained, compared with the control group statistically significant difference (P<0.05),and has effects,the relationship between the aging.Conclusion; Modified Yinshao pulvis inhibits HSC activation, The mechanism of anti hepatic fibrosis may be related to the down-regulation the expression of TGF-beta 1 and Smad3, and blocks TGF-beta 1/Smad signaling pathway.
