| Objective:To observe the effects of KangXingYing (KXY),a kind of traditional Chinese medicine,on the rat’s of ischemic cardiomyopathy(ICM),to explore the underlying mechanisms of myocardial fibrosis, and to broaden the use of the decoction with warming yang for diuresis and promoting blood circulation for removing blood stasis.Methods:Forty-eight male SD rats,besides of eight rats to following sham-operated groups, preparation of ICM in the rat model assigned to the following three groups randomly;model group,KangXinYingroup(KXY group),and the control group (captopril group),for each group eight rats.The sham-operated group,the left anterior descending coronary artery only thread ligation after thoracotomy,and others conduct the left anterior descending coronary artery ligation. Electrocardiogram showed ST segment elevation and (or) T wave towering change or inversion in Lead Ⅱ,which indicated acute myocardial infarction (AMI).At the fifth week after AMI, KXY groups starts the lavage medication with of KXY in morning and evening,For captopril group,it starts the lavage medication with captopril in morning and evening.Fifth weeks after operation,control group and model group starts the lavage medication with 0.9% saline in lml/100g body weight each time.After two weeks of treatment, heart was harvested.HE staining and Immunohistochemistry were used to detect the histological changes,Sirius Red F3B in saturated carbazotic acid staining was used to employed to calculate the content of collagen,Western Blot were used to detected the protein content of the MMP-2,MMP-9 and TIMP-2,QPCR detect the expression of MMP-2, MMP-9 and TIMP-2 mRNA.Results:(1)HE staining result:Compared with sham-operated group, hyperplasia of myocardi-al fibers,Horizontal stripes blur or fault,focal myocardial necrosis,disarrangement of my-ofibers,degeneration of cardiomyocates, edema, the nucleus concentration in model group, there are inflammatory cell infiltration, such as macrophage,nuclear enrichment.Compared with model group,A little of degeneration of cardiomyocates edema.(2)Sirius Red in saturated carbazotic acid staining:Compared with sham-operated group, myocardial interstitial collagen fibers was significantly increased in model group;Compared with model group, Captopril group and KXY group Myocardial fibrosis decreased significantly.(3)Immunohistochemistry detection:Compared with sham-operated group,in model group,myocardial tissue the expression of MMP-2 and MMP-9 increased significantly (P<0.01),TIMP-2 quantity decreased(P<0.05).Compared with model group,myocardial tissue the expression of MMP-2 and MMP-9 quantity decreased(P<0.01),TIMP-2 increased (P<0.05). Captopril group and KXF group has no statistical significance(P>0.05).(4)Real-time Quantitative polymerase chain reaction:Compared with control group,MM P-2 and MMP-9 mRNA increased significantly (P<0.01) TIMP-2 mRNA expression qua-ntity decreased in model group(P<0.05);compared with model group,TIMP-2 mRNA exp-ression quantity increased in KXF group and captopril group(P<0.05),MMP-2 and MMP-9 mRNA quantity decreased(P<0.01).(5) Western Blot detection:compared with sham-operated group,protein expression quantity of MMP-2 and MMP-9 in model group increased (P<0.01),and the TIMP-2 protein expression quantity decreased(P<0.01).compared with model group,the MMP-2 and MMP-9 protein expression quantity decreased(P<0.01or P<0.05),in KXF group and captopril group,the TIMP-2 protein expression expression quantity increased(P<0.05).KXY of MMP-2,MMP-9 and TIMP-2 mRNA expression and its effect on the protein expression trend about the same.Conclusions:(1) KXF can Relieve edema of myocardial cell degeneration necrosis and interstitial collagen fiber hyperplasia,which indicated that KXY can alleviate myocardial fibrosis of ICM.(2) KXF can reduce myocardial fibrosis of ICM,suppressed the expreesion of MMP-2 and MMP-9,and promoted the expression of TIMP-2. |
