Clinical Study Of Epithelial Ovarian Cancer In Vitro Drug Susceptibility Testing

Objective:To investigate the use of ATP bioluminescence assay of epithelial ovarian cancer drug sensitivity test for guiding the feasibility of postoperative chemotherapy.Methods: June 2013 to October 2015, the First Affiliated Hospital of Soochow University Obstetrics and Gynecology 30 treated cases of ovarian cancer,tumor tissue samples were collected under sterile conditions surgery.Choose eight groups of chemotherapy drugs Including TAX+DDP 、 TAX+CBP 、 TAX 、 Topotecan+DDP 、DDP+CTX+ADM、DDP+ADM、GEM+CBP、Pemetrexeddisodium and set different drug concentration Including 200%PPC(plasma peak concentration)、100%PPC、50%PPC、25%PPC、12.5%PPC, and co-cultured with tumor cell. Using ATP bioluminescence assay to obtain the highest inhibition rate to the set of chemotherapy, According to the response toDetermine the chemotherapy after surgery, compared to 30 people who did postoperative empirical chemotherapy at the same period, Follow-up of serum CA125 in two groups of patients return to normal time, chemotherapy treatments, progression-free survival and side effects.Results: the chemosensitivities priority of epithelial ovarian cancer to the anticancer drugs in plasma peak concentration:TAX+DDP>TAX+CBP>TAX> Topotecan +DDP > DDP+CTX+ADM > DDP+ADM > GEM+CBP >Pemetrexeddisodium. There was no statistical difference in age, stage, pathological type, postoperative residual lesion in the two groups of patients. Susceptibility of clinical effectiveness was higher than another group, there was differences between the two groups of the recent clinical curative effect(χ2=6.121,P<0.05). Two groups of patients with serum CA125 returned to normal time, chemotherapy regimen, progression-free survival, the cases of adverse reactions and degree is not statistically significant.Conclusion: this study demonstrates different patients with different chemotherapy drugs sensitivity, and a significant individual differences. Tumors in vitro susceptibility test results to guide the clinical curative effect of patients with clinical chemotherapy and has a good correlation, to guide individualized chemotherapy postoperatively in patients with ovarian epithelial carcinoma has important clinical value.
