Objective: To detect the expression of NCOA5 in gastric cancer and to analyze the relationship between the expression of NCOA5 and the clinicopathological parameters of patients. To study the effect of abnormal NCOA5 on cell proliferation and migration of human gastric cancer cell line SGC7901.Methods: The tissue samples were collected from 40 patients with gastric cancer, and the expression of NCOA5 was detected by immunohistochemistry. The lentiviral vectors with specific interference NCOA5 sequences and a random sequence as a control were constructed. Lentivirus was packed in 293 T cells and then infected the gastric cancer cell lines SGC7901. Overexpression of NCOA5 inSGC7901 cells was also carried out by lentivirus infection. Western blot was used to detect the expression of NCOA5, MTT and wound-healing assay were used to analyze the cell proliferation and migration.Results: The expression of NCOA5 in gastric cancer tissue samples was notably higher than that in the adjacent normal specimens. And it was found to be significantly correlated with the tumor invasion depth, lymph node metastasis and TNM stage(P<0.005). The gastric cancer cell line SGC7901 infected with interference NCOA5 lentivirus showed efficiently reduced level of endogenous NCOA5 expression and significantly decreased cell proliferation and migration while the cell infected with overexpression NCOA5 lentivirus showed the opposite results(P<0.005).Conclusions:(1) NCOA5 is highly expressed in gastric cancer tissues and the positive expression rate of NCOA5 correlates with tumor invasion depth, lymph node metastasis and TNM stage;(2)NCOA5 can promote the ability of proliferation and migration in the gastric cancer cell line SGC7910;(3)To compare the expression of NCOA5 in gastric cancer and its effect on the function of gastric cancer cells, suggesting that NCOA5 may be the pathogenesis related genes of gastric cancer, which may provide reference for early diagnosis of gastric cancer.
