ObjectiveThrough random control group catgut embedding stress control study for five Zang Shu acupoint catgut embedding anti-aging and anti stress damage role to provide laboratory evidence.MethodsThe 3 month old SD male rats (n= 60), SPF randomly divided into blank group, model group, model the catgut embedding group, model group of stress, stress model of catgut embedding group,12 in each group. The blank control group (A) for 8 weeks by intraperitoneal injection, but not stress and Catgut embedding. Model group (B) was given 8 weeks by intraperitoneal injection of D-galactose, but no stress. Model embedding group (C) in (B) on the basis of acupoint catgut embedding, from the beginning of the first week, the 1 time in 1 weeks, a total of 8 times. Model stress group (D) was administered 8 weeks by intraperitoneal injection of D-galactose, starting from fifth weeks to brake stress, once a day, a total of 4 weeks of stress. The stress model (E) in the embedding group (D) on the basis of acupoint catgut embedding, from the beginning of the first week, the 1 time in 1 weeks, a total of 8 times. Method of stress is to build model rats were exposed to immobilization stress device to completely restrict the activity of rats of,4 weeks of continuous stress and by sequential extended braking time increase the stress intensity. All rats were divided into eight weeks, embedding the selected five viscera Shu points are a reference to the "experimental acupuncture" (edited by Li Zhongren,2003 China Press of traditional Chinese Medicine). The general state of the rats weight, hair, mental and eating of the morphological changes were observed in model rats before and after, before and after stress and catgut embedding therapy before and after the three time points. On the day before and after the experiment, all rats were exposed to the open (open-field) test, and the behavior of the rats was measured. The behavioral test was performed immediately after the completion of all the rats, the rats were taken from the hippocampus, the hippocampus was measured by DA, acetylcholine Ach, monoamine oxidase MAO and protein content. The results were analyzed by statistical software SPSS20.0.Results1、Weight dataComparison between groups:each group had no significant difference in body weight data of first days (P=0.515), the weight data of twenty-eighth days (P=0.000) and fifty-sixth day weight data (P=0.000) with statistical significance.Group comparison:in twenty-eighth days and fifty-sixth days A (P=0.480), B (P=0.650), C (P=0.170) group had no statistically significant difference, in first days, fifty-sixth days D group had no significant statistical difference (P=0.724), the rats in the group weight data of the other time points were statistically the difference (P< 0.05).2、Autonomous behaviorThe central region of the time of the rats (P=0.000) and the central region distance data (P=0.000) had significant difference.The results show that stress can cause the body weight loss and accelerated aging; catgut embedding on body aging can play a certain effect.3、The content of the organizationThe dopamine content in hippocampus of rats in each group (P=0.000), the content of acetylcholine (P=0.000), protein content (P=0.000) and the content of monoamine oxidase (P=0.000) had significant difference.The results show that stress can reduce the content of neurotransmitter in the hippocampus of the brain, thus speeding up the aging; catgut embedding neurotransmitters in the hippocampus of the brain to reduce to a certain extent.ConclusionBy embedding control experiments and in the hippocampus of the brain, dopamine, acetylcholine, monoamine oxidase, protein content, five Zang Shu acupoint catgut embedding on anti-aging and anti stress injury is effective in the study of rat morphology and autonomous behavior.
