Objectiveatherosclerosis is a pathological basis of many cardiovascular and cerebrovascular diseases. With the improvement of living standards, atherosclerosis has been the column of the major causes of death. Therefore, to prevent the occurrence and development of atherosclerosis, which has important significance for cardiovascular and cerebrovascular diseases. In recent years, a large number of chinese herbal monomer in the prevention and treatment of atherosclerosis are found, and resveratrol is one of these. Resveratrol is expected to provide further clues to improve the clinical status of patients.Resveratrol, a polyphenolic naturally antioxidant, has been a research focus on the preventment and treatment. In the preliminary works, we have confirmed that resveratrol may indeed have prevented the atheroscularsis. There has substantial evidence that nitric oxide and nitric oxide synthase are closely related to atheroscularsis.It is reported that estrogen plays an important role in the atheroscularsis. Therefore, we explore the interrelation with estrogen receptor and nitric oxide synthase in the process of resveratrol inhibiting atherosclerosis.Methods1. The role of resveratrol in atherosclerosisAtherosclerosis model is successfully constructed by C57 mice ovariectomizing (OVX) and high-fat diet. We verify the model and the role of resveratrol in atherosclerosis through the four serum lipids detection and immunohistochemistry.2. The influence of nitric oxide and oxidative stress in the process of resveratrol in atherosclerosisWe measure the nitric oxide and expression of nitric oxide synthase and superoxide anion in aorta, through immunohistochemistry and western blot.3. The influence of estrogen receptor in the process of resveratrol in atherosclerosisWe measure the expression of estrogen receptor in aorta through immunohistochemistry and western blot, to observe the relationship among the estrogen receptor, resveratrol and atherosclerosisResults1. The blood-lipoids and Lipidoses of OVX and high-diet mice increased distinctively. So the atherosclerosis model is sucessful. The blood-lipoids and Lipidoses of mice in resveratrol group is lower distinctively. It explain that resveratrol can inhibit the atherosclerosis.2. The nitric oxide and superoxide anion of OVX and high-diet mice are more than sham group. The nitric oxide and superoxide anion of mice in resveratrol group is lower distinctively. The expression of inducible nitric oxide synthase in resveratrol group is lower than ovx and high-diet mice, however, the expression of endothelial nitric oxide synthase in resveratrol group goes up. It explains that the inhibition of resveratrol in atherosclerosis may happen through the regulation of nitric oxide and oxidative stress.3. The expression of estrogen receptor in OVX and high-diet mice is less than sham group, but the expression in resveratrol group is more than the OVX and high-diet group. It explains that resveratrol inhibits the atherosclerosis through the adjustment of estrogen receptor.ConclusionResveratrol can raise the expression of estrogen receptor, inhibit the expression of inducible nitric oxide synthase, and increase the expression of endothelial nitric oxide synthase, and reduce the nitric oxide, restrain the oxidative stress, to inhibit the atherosclerosis.
