BackgroundAlzheimer’s disease (Alzheimer’s Disease, AD), is the most common cause of dementia, known as primary clinical Alzheimer’s disease, is a common insidious onset of sexual development of degenerative disease of the central nervous system, the patient may be due to secondary infections and complications and death. AD pathogenesis complex, cause unknown, As human society entered the aging accelerated pace, the number of elderly population aged over 65 increased year by year, studies show that the number of dementia patients in China has more than 8 million by 2040 will be over 26 million. Dementia high morbidity, will give family and society a heavy burden. Therefore, prevention and treatment of Alzheimer’s disease (AD) has a positive social significance, Chinese medicine has a unique position and broad prospects in the prevention and treatment of AD. Situation with explicit Alzheimer’s disease, to elucidate the pathogenesis, the discovery of new drug targets, to find effective drug treatment for Alzheimer’s disease.Protocatechuic acid (Protocatechuic acid, PCA) is found in many vegetables and fruits in a natural phenolic compounds, and the active ingredient is a lot of Chinese medicine, such as treating diabetes, salvia, hibiscus and so on. And treating diabetes is one of the "four South medicine" of pharmacological study found that having cancer, cardiovascular protection and so on, on nerve cells has anti-aging, anti-oxidation, reduce excitotoxicity and so on [4]. The study found that the main component of treating diabetes protocatechuate good neuroprotective effect. On the pharmacological effects of protocatechuic it has been numerous reports, protocatechuic acid has anti-inflammatory, antioxidant, inhibiting tumor; protocatechuic ischemia, hypoxia neuronal cells have a protective effect; protocatechuic acid significantly inhibited MPP+, H202 or rotenone-induced PC12 cell apoptosis and oxidative stress, has a good antioxidant and neuroprotective effectsObjectiveThis study in PC12 cells induced by A β1-42 deposition AD cell model to study the protective effect and mechanism of protocatechuic A β1-42 oligomer-induced toxicity in PC12 cells, in order to better define protocatechuic neuroprotective effect and mechanism of action, the further development of effective treating diabetes and its original monomers catechin and provide experimental evidence.MethodFirst identified by immunofluorescence fiber A β1-42 oligomers, the A β1-42 of PC12 cells and at different time points (0,3,6,9,12,24 h) Morphology of PC12 cells, using MTT colorimetric assay to determine cell viability modeling conditions. When protocatechuic acid under conditions of 24 h of 200 u mol/L, the concentration on cell viability increased. At the same time, Western-blot results showed that protocatechuic acid increased the expression level of Beclin1.ConclusionProtocatechuic acid can significantly protect the Aβ1-42 induced PC12 cells, PC12 cells and reduced Aβ1-42 toxicity, its mechanism may be related to the level of autophagy for its anti-AD acts deeper research value.
