ObjectiveAfter abdominal surgery, gastrointestinal dysfunction often occurs, resulting in a series of clinical symptoms. In the clinical study of promoting the recovery of gastrointestinal function after operation, a large number of clinical studies indicate that acupuncture can reduce the degree of postoperative gastrointestinal dysfunction, promote gastrointestinal function recovery, and is currently widely used in clinical practice. Through the preliminary study, it is found that the recovery of gastrointestinal function may be closely related to the regeneration of ICCs in the gastrointestinal tract. CO gas signaling pathway is the ICCs repair and regeneration process in an important way, and CO is mediated by an inhibitory neurotransmitter gastrointestinal smooth muscle relaxation, when the neuron is stimulated, Ho co synthesis and release, mediated by nerve conduction. In this study, we observed the acupuncture of "Jin" group "foot three needle" effect on SD rat gastrointestinal tract CO gas signaling pathway after colonic anastomosis to explore acupuncture "foot three needle" mechanism of gastrointestinal dysfunction in SD rat gastrointestinal function recovery after operation.MethodThe SD rats were randomly divided into 3 groups:normal group, model group, acupuncture group, model group and acupuncture group were divided into 3 days group,5 day group and 10 day group according to the different intervention time.Model group:the model of colonic anastomosis was made. After anesthesia along the linea alba in the lower abdomen cut open the stomach, and the lower end of cecum 2cm cut, suture of colon, followed by suture of peritoneum, muscle and skin, postoperative not acupuncture.Acupuncture group:the model group and model group were given acupuncture intervention according to the intervention time.Small intestinal propulsion rate measurements:SD rats before samples were obtained, by irrigation stomach decimator lml ink of rats by intragastric administration, filling the stomach 30min after they were killed by cervical dislocation and tissue scissors to open the abdomen, fully exposed gut, carefully stripping large intestine, small intestine, a measurement distance of the full length of the small intestine and the pylorus to ink distal. According to the formula:small intestine propulsion rate= (ink travel length /total length of small intestine)*100%, calculate the small intestinal propulsion rate of each rat.CO and HO-1 and HO-2 content determination:after the SD rats were sacrificed by dislocation and interception on the lower end of cecum anastomotic about 1 cm long, the intestinal tissue clean, along the longitudinal axis of the divided into three parts, respectively, for to detect HO-1, HO-2, CO and electron microscope were used to observe; in accordance with specimens of different processing method of the intestinal tissue pre-treatment. The HO-1, HO-2 activity and CO content were determined by dual wavelength spectrophotometer. The ultrastructure of Cajal cells was observed by electron microscope.Result1. Small intestinal propulsion rate determination results:compared with the normal group, model group of 3 days,5 days, model group, model group of 10 days,3 day acupuncture group the results were statistically significant (P<0.01), suggesting that the normal group has intestinal propulsion rate higher than the 4 group; compared with the normal group, the acupuncture group 5 days and 10 days the acupuncture group was not statistically significant, suggesting that normal intestinal propulsive rate and 5 days 10 days acupuncture group, acupuncture group had no significant difference; comparison between groups:3 day acupuncture acupuncture group was significantly lower than the rate of small intestine 5 days acupuncture group and acupuncture group (P<0.01) 10 days,5 day acupuncture group, intestinal propulsive rate 10 days of acupuncture and group the results were not statistically significant between the two groups, suggesting that small intestinal propulsion rate have no significant difference; comparison between groups:model group, model group of 3 days and 5 days compared to the model group the results were not statistically significant, model group of 5 days and 10 days Model group compared the results were statistically significant (P<0.05); compared with the model group, acupuncture group,3 days acupuncture group and 10 day model group was not statistically significant difference (P>0.05).2. HO-1 assay results:compared with the normal group, model group of 3 days,5 days, model group, model group of 10 days,3 day acupuncture group the results were statistically significant (P<0.01),5 day 10 day acupuncture group, acupuncture group the results were not statistically significant (P>0.05); acupuncture group between groups, with 3 days of acupuncture compared with the 5 day acupuncture group, the results were not statistically significant (P>0.05),10 day acupuncture group results were statistically significant (P<0.01); the 5 day acupuncture group and acupuncture group compared the results of 10 days was not statistically significant (P>0.05); comparison between groups:model group and model group of 3 days compared to model group of 5 days no statistical significance (P>0.05), model group of 10 days the results were statistically significant (P<0.01), model group of 5 days and 10 days compared to the model group, the results were not statistically significant (P>0.05); compared with the model group and the acupuncture group and the model group:3 days, acupuncture group acupuncture group of 3 days,5 days,10 day acupuncture group results have statistical significance (P<0.01); compared with the model group of 5 days,3 days acupuncture group results without statistical significance (P>0.05), acupuncture group of 5 days,10 day acupuncture group results have statistical significance (P<0.01); and 10 days compared to the model group, acupuncture group of 3 days,5 days acupuncture group results without statistical significance (P>0.05), acupuncture group of 10 days results have statistical significance (P<0.05).3. Results for the determination of activity of HO-2:group and normal compared group acupuncture acupuncture group of model group of 3 days, the model group, model group of 5 days,10 days,3 days,5 days,10 day acupuncture group results had no statistical significance (P>0.05); acupuncture group comparison showed no statistical significance (P>0.05); model between groups comparison showed no statistical significance (P>0.05); acupuncture group and model group comparison showed no statistical significance (P>0.05)4. The determination results of CO content:compared with the normal group, model group of 3 days,5 days, model group, model group of 10 days,3 days and 5 days of acupuncture group, acupuncture group the results were statistically significant (P<0.01),10 day acupuncture group, the results were not statistically significant (P>0.05); acupuncture group between groups, acupuncture group 3 days and 5 days of acupuncture, acupuncture group, acupuncture group 10 days the results were statistically significant (P<0.01), 5 day 10 day acupuncture group and acupuncture group the results were statistically significant (P<0.01); model group between groups, compared with the model group of 3 days,5 days model group, the results were not statistically significant (P>0.05). The 10 day model group results were statistically significant (P<0.01); compared with the model group and the acupuncture group, compared with the model group of 3 days,3 days and 5 day acupuncture group, acupuncture group, acupuncture group 10 days and the results were statistically significant (P<0.01); compared with the model group of 5 days,3 day acupuncture group Results there was no statistical significance (P>0.05), acupuncture group of 5 days,10 day acupuncture group results have statistical significance (P<0.01); and 10 days compared to the model group, acupuncture group of 3 days,10 days acupuncture group results were significant (P<0.01),5 day acupuncture group showed no statistical significance (P>0.05)5. Electron microscope Cajal cell results:(1). Normal group:interstitial cells of Cajal into fusiform, nuclei were clear, oval, chromatin in the cells of uniform, clear, around the nucleus and little cytoplasm, intracellular organelles such as well-developed mitochondria, endoplasmic reticulum and Golgi apparatus; clear cytoplasmic intermediate filament, large number of distributed at the periphery of the nucleus. The basement membrane integrity; synaptic connection clear.(2). Model group:CDModel 3 days group:interstitial cells of Cajal raised significantly reduced, less cytoplasm and cytoplasm appeared a large number of vacuoles, significantly reduced the number of organelles, cells exist extensive lesions, mitochondrial swelling, mitochondrial cristae; Endocytoplasmic reticulum, endoplasmic reticulum degranulation; basement membrane is incomplete,day model group:Cajal cells were irregular in shape, karyopyknosis, heterochromatin margination, intracellular cytoplasmic slightly rich, cell structure slightly clearer; swelling of mitochondria, unclear structure; intermediate filament number slightly, basement membrane incomplete.③10 day model group:cells with abundant cytoplasm, large number of organelles, part of the structure is not clear; mitochondria swelling, visible a few lysosomes; part of the rough endoplasmic reticulum degranulation, clear intermediate filaments, basement membrane slightly complete.(3). Acupuncture group:①Acupuncture 3 days group:the cell cytoplasm is rich, the number of organelles is more, the structure is more clear; mitochondria part of swelling; endoplasmic reticulum expansion is not obvious; the intermediate filament is clear, the basement membrane is slightly complete.②5 day acupuncture group:interstitial cells of Cajal in regular shape, fusiform, chromatin is homogeneous, perinuclear cytoplasm rich, large number of organelles, clear structure, more mitochondria, clear structure; no expansion of endoplasmic reticulum; visible a few Golgi bodies and lysosomes; basement membrane complete.③10 day acupuncture group:interstitial cells of Cajal into fusiform, nuclei were clear, oval, chromatin in the cells of uniform, clear, around the nucleus and little cytoplasm and cytoplasmic organelles well developed; clear cytoplasmic intermediate filament, large number of distributed at the periphery of the nucleus; basement membrane; synaptic clearly link.conclusion1. acupuncture can improve the small intestinal propulsion rate of gastrointestinal dysfunction in SD rats, the recovery time of intestinal peristalsis in acupuncture group was significantly shorter than that in model group.2. acupuncture can reduce the stress of the gastrointestinal tract H0-1, HO-2 activity, so that the amount of CO synthesis decreased, CO mediated gastrointestinal smooth muscle relaxation function weakened, and promote the recovery of gastrointestinal motility function.3. acupuncture has a moderating effect on the HO-2 activity, which can promote the recovery of the injury caused by surgical injury.4. in POI, the high concentration of CO and KIT expression decreased, suggesting that CO is not conducive to the high concentration of ICC survival...
