ObjectiveHeart failure refers to the heart in has circumfluence of blood of right amount of cases, unable to maintain adequate cardiac output, it lead to the amount of tissue perfusion decreased, circulatory disturbance syndrome. The heart failure is a common outcome of many cardiovascular diseases. Such as primary myocardial damage, ischemic cardiomyopathy, myocarditis and dilated cardiomyopathy, hypertension, aortic stenosis of pressure overload and heart valve closure insufficiency, the capacity of the left and right ventricular shunts, these factors can lead to heart failure. When the myocardial contraction force reduced, in order to guarantee the normal cardiac output the body began to appear compensatory mechanism, like Frank-Starling mechanism, myocardial hypertrophy, neurohumoral compensatory mechanism. Current detecting of heart failure rely on heart ejection fraction, the BNP, X-ray film, etc. Nearly two years found that the role of lncRNA gene is not only the noise, can also be encoded protein, secreted into the blood, proved to be related to heart failure, and can be combined for treatment of heart failure. This paper involves lncRNABC088254 which has be pointed out that was the key of the myocardial cell energy. The expression level of lncRNABC088254 may be associated with the degree of heart failure. It is urge to explore more sensitive index of heart failure. And then the heart failure can be prevention.Methods1) We chose 6 heart tissue of heart failure caused by dilated cardiomyopathy from shandong province hospital,4 cases tissue of normal cardiac function as the control group. We analyzed age, gender by statistics. The analysis of age was taken the way of independent sample t-test analysis; the analysis of gender was taken the way of Chi square test. The values of analysis results were expressed with P, P<0.05 or P<0.01 is statistically significant.2) Applying the myocardial tissue to test expression of lncRNABC088254, the results confirmed that lncBC088254 was expressed in heart failure. While testing lncRNABC088254 genetic level in patients, we analyzed the data of left ventricular ejection fraction and BNP. The purpose to prove the relationship between lncRNABC088254 expression in heart failure disease, left ventricular ejection fraction and the BNP’s value were statistically significant. The results are presented as the mean±SEM. Comparisons between groups were conducted using one-way analysis of variance (ANOVA) followed by Bonferroni’s post hoc test. P<0.05 and P<0.01 were considered to indicate statistically significant and highly significant differences, respectively, for all of the analyses. All of the statistical analyses were conducted with the SPSS 17.0 statistical software package (SPSS, Inc., Chicago, IL, USA).3) lncRNABC088254 in vitro, through the homologous transcription to find the lncRNABC088254 sequence of the rats, stimulate myocyte hypertrophy with angiotensin Ⅱ and detect lncRNABC088254 expression level of myocyte hypertrophy compared with the control group; Using Smart Silencer (a kind of siRNA, specificity of RNA interference agent) to obstruct the expression of lncRNABC088254 and detected the expression of phb2 by RT-PCR and Western blot.Results1) In view of the patient’s age, gender factors were analyzed, and the analysis results show that the distribution of age was described using the mean+/- standard and the difference was no statistically significant (P> 0.05); The analysis of gender with four card party inspection to table, the final results show that the differences have no significance (P>0.05). Through the statistical analysis of the age and gender we can think that it doesn’t make the influence of the result of the experiment.2) We can extract RNA by myocardial tissue of Patients. The results of RT-PCR showed the expression of lncRNABC088254 in heart failure tissue was raised compared with the level of the normal cardiac function of valvular replacement papillary muscle. The difference was statistically significant (P<0.01). We also found that the IVSd of experimental group compared with the control group significantly increased, but the left ventricular ejection fraction was lower compared with the normal control group (P<0.05); the levels of BNP in experimental group was expressed in high volume and the difference was statistically significant (P<0.01); Experimentalthe expression of the BNP, lncRNABC088254 in the control group were significantly increased (P<0.05)3) In vitro model of myocyte hypertrophy, the RT-PCR results show that the expression level of lncRNABC088254 and related gene phb2 were rose compared with the control group under the stimulus of angiotensin Ⅱ. The difference was statistically significant (P<0.01); with lncRNABC088254 interference, the results showed that the expression level of lncRNABC088254 was lower than the negative control group and blank control group, phb2 gene expression level was lower than the negative control group and the blank control group. The difference was statistical significance (P<0.05). The results of protein gel electrophoresis showed that the protein of phb2 grey value is lower than the negative control group and blank control group. The differences was statistically significant (P<0.05).ConclusionUsing randomized controlled methods, this study analyzes the effect of lncRNABC088254 in heart failure disease was very important in the course of disease, heart failure. The effect of lncBC088254 was an iconic factor in heart failure. The results provide a new way of clinical heart failure diagnosis, and provide the theoretical basis for the role of lncRNA.
