BackgroundPituitary adenomas are one of common intracranial tumors, and about 40 percentage of which invade surrounding structures and show aggressive behavior. We define them as invasive pituitary tumors. Because of the violation of important tissues around the saddle, the postoperative recurrence and the effect of endocrine changes, the prognosis is poor. it is difficult to be completely removed in operation. In recent years, the diagnosis, the development mechanism and molecular biological therapy of invasive pituitary adenomas have been hot topics. β3GnT8 is a new member of β3GnTs family. Previous studies found that β3GnT8 can promote the synthesis of polylactosamine on the branch of N-glycans which was closely related to the invasion and metastasis in tumor cells. CD147 is a tumor-associated antigen which was N-glycosylated and contained a series of complex polylactosamine on the branch of N-glycans that plays a key regulatory role for MMP activities. Many studies have reported that the expression of β3GnT8 in various tumor cells, but its expression in pituitary tumor is rarely reported. In this study, we will study its expression in pituitary tumors, and its impact on the invasive behavior, in order to better guide the diagnosis, treatment and prognosis of pituitary tumors.This experiment is divided into two parts.First Part:Eexpression of β3GnT8 in invasive pituitary adenomas and noninvasive pituitary adenomasObjective To explore the expression of β3GnT8 in invasive pituitary adenomas and noninvasive pituitary adenomas.Methods A total of 58 cases of pituitary adenoma were collected from patients who underwent surgery in Shandong Province Hospital from September,2013 to February,2015, including 26 males and 32 females, with an average of 38.2 years old. Among them, there were 16 cases of growth hormone secreting pituitary adenoma,17 cases of prolactin secreting pituitary adenoma,3 cases of adrenal cortical hormone secreting pituitary adenoma,19 cases of non functional pituitary adenoma,3 cases of mixed pituitary adenoma. According to the criteria for invasive pituitary adenomas, they are divided into two groups(32 cases in invasive group and 26 cases in non-invasive group). Each sample were divided into two parts. One part was used to determine the expression of β3GnT8 by immunohistochemical method.Results The β3GnT8 expression rate was significantly higher in the invasive group than in the non-invasive group (87.5% VS 57.5%, P< 0.05).Conclusion The β3GnT8 expression in the invasive group is higher than in the non-invasive group and might relate with the aggressiveness and invasion of pituitary adenomas. Second Part:Relation of expression of β3GnT8, HG-CD147 and MMP-2 in pituitary adenomasObjective To explore the relation of expression of β3GnT8, HG-CD147 and MMP-2 in pituitary adenomas.Methods The other part of the samples were used to determine the expression of β3GnT8, CD147 and MMP-2 by Western blotting. After comparing each relation among those three parameters, we could have a primary knowledge about the influence of β3GnT8, CD147 and MMP-2 on the invasion of pituitary adenomas.Results Western blotting revealed that the expression of β3GnT8, HG-CD147 and MMP-2 in the invasive group was high compared with the non-invasive group (t=2.72, P< 0.05;t=2.81, P< 0.05;t=2.68, P< 0.05), and as the expression of β3GnT8 raising, the expression of HG-CD147 and MMP-2 also increased. Pearson correlation showed there were mutually positive correlations among β3GnT8, CD147 and MMP-2 expressions in invasive pituitary adenomas (r=0.78, P< 0.05; r=0.64, P<0.05; r=0.52, P<0.05)Conclusion The β3GnT8 expression might relate with the aggressiveness and invasion of pituitary adenomas, it may be an useful marker for predicting invasiveness and provides a new way for the diagnosis and treatment of invasive pituitary adenomas.
