Background and objective:Diseases of Enteric Nervous System have been a hard obsession all the time. There are lots of influencing factors and varied pattern of manifestation. Their causes are not clear and definite, further more, morbidity of Enteric Nervous System diseases show a increasing trend. Thought a part of Enteric Nervous System diseases could be cured by surgical operation, their postoperative complications and postoperative recovery is influenced by lots of unascertained factors, and these are our task of thid study. Neuroligin 1 exist in body of human and other mammals, and is expressed in ENS. Meanwhile, it play a important role in ENS. Our aim is to research and investigate the expression of Neuroligin 1 in ENS of Wistar rat and discuss relationship between Neuroligin 1 and ENS or ENS diseases.Methods Collect 30 embryonic rats’ intestinal tissues from the 16th day pregnancy(E16),the 18th day pregnancy(E18), the 20th day pregnancy(E20) and born on the first day (B1). In the light of experiment mthods of Double-immunofluorescent labeling, Western blot, Real-time Quantitative polymerase chain reaction (qRT-PCR), ELISA and immunohistochemistry, studying the expression of Neuroligin 1 in each period.Results In the four periods:E16, E18, E20 and B1, Double-immunofluorescent labeling showed that the average optical density of positive area were E16(0.046± 0.031), E18(0.146±0.033), E20(0.233±0.034), B 1(0.345±0.344), meanwhile, their differences were significant difference(F=441.614, P=0.000, P<0.05). Meanwhile, compare in the four groups, there are differences in them and their differences were statistical significance(P=0.000, P<0.05). Western blot displayed that the average protein expression were E16(0.053±0.025), E18(0.146±0.024), E20(0.249±0.034), B1 (0.533± 0.049), and their differences were statistical significance(F=1070.296, P=0.000, P<0.05). At the same time, compare in the four groups, there are differences in them and their differences were statistical significance(P=0.000, P<0.05). qRT-PCR indicated that the comparative mean concentrations of mRNA were E16(2.543±1.294), E18(4.637±1.830), E20(6.814±1.552), B1(8.345±1.364) and their differences were statistical significance(F=81.440,p=0.000, P<0.05). At the same time, compare in the four groups, there are differences in them and their differences were statistical significance(P=0.000, P<0.05).ELISA displayed that the average protein expression were E16(0.069±0.061), E18(0.153±0.102), E20(1.087±0.093), B1(1.678±0.085) and their differences were statistical significance(F=2396.068, p=0.000, P<0.05). At the same time, compare in the four groups, there are differences in them and their differences were statistical significance(P=0.000, P<0.05). Immunohistochemistry displayed that the average protein expression were E16 (±~+),E18 (±~++),E20 (++~+++), B1 (+++~++++) and their order was E16<E18<E20<B1. Expression trends of Neuroligin1 in Immunofluorescence technique, Western blot, Real-time Quantitative polymerase chain reaction (qRT-PCR), ELISA and immunohistochemistry were the same.Conclusion During the period from E6 to B1, Neuroligin1 increased gradually along with Wistar rat ENS’s growth. The earlier Neuroligin1 abnormal expression occur, the more serious correlative diseases were. ENS diseases which were related to Neuroligin1 were congenital diseases probably.
