| Surgery is an important treatment for gastric cancer. But to the patients with locally advanced gastric cancer, the prognosis of surgical resection alone is not satisfactory. The recurrence rate of simple surgical treatment to gastric cancer was 60%. It is as high as 80% to the patients with lymphatic metastasis. To actively expand the scope of operation, including spleen and retinal resection, positive lymph node dissection, can’t effectively improve the survival rate of patients with gastric cancer or reduce the recurrence rate. Therefore, the comprehensive treatmen combined with surgery, radiotherapy and chemotherapy is the current clinical treatment of gastric cancer. However, the patients of gastric cancer have greater individual differences. Their response to treatment is not the same, and the treatment effect is uneven. It’s important significance for reducing the side effects of the therapy, improving the treatment effect, prolonging the survival time of patients to looking for suitable molecular markers for individual treatment of patients with gastric cancer.Ribonucleotide Reductase Large Subunit M1 has been regarded as a kind of anti-oncogene. RRM1 can enhance the monitoring function of G2 cells and promote to repair and apoptosis after DNA damage. The Topoisomerase II alpha is an important hydrolase or synthase. It plays an important role in DNA replication, chromosome segregation, chromatin condensation and gene expression and other biological processes.Objective:The study aimed at investigating the expression of RRM1 and TOP2a in 88 cases of gastric cancer and 20 cases of normal tissue by immunohistochemical method. To analyze the relationship between the expression level and patient’s age, gender, depth of tumor infiltration depth, lymph node metastasis, differentiation degree, clinical stage or distant metastasis by statistical methods. And to analyze the relationship between the expression of RRMl and TOP2a.Methods:The first people’s Hospital of Jingzhou, Selected 88 cases of paraffin specimens of gastric adenocarcinoma that was Removed by surgery between May 2014 to November 2015 in The first people’s Hospital of Jingzhou and were diagnosed two pathology Professor independently. And 20 normal gastric tissues were selected as control group. Investigate the expression of RRM1 and TOP2a in the specimens. To analyze the relationship between the expression level and patient’s age, gender, depth of tumor infiltration depth, lymph node metastasis, differentiation degree, clinical stage or distant metastasis by SPSS 19.0.And to analyze the relationship between the expression of RRM1 and TOP2a.Results:The positive expression rate of RRM1 in gastric adenocarcinoma was 83.0%(73/88). And RRM1 was negative in the entire normal gastric tissues. The positive expression of RRM1 was not associated with patient’s age (x2=0.352, P=0.553), gender (x2=0.493, P=0.482), depth of tumor infiltration depth (x2=0.007, P=0.933), lymph nodes metastasis (x2=0.121, P=0.728) and distant metastasis (P=0.415). But it was related to the degree of tumor differentiation (x2=7.740, P=0.021) and clinical stage (x2=5.733, P=0.017). The stages of gastric cancer was later, the expression of RRM1 in gastric cancer tissues was higher.The stage of tumor was earlier, the expression of RRM1 was lower. The degree of tumor differentiation was low, the level of RRM1 expression was higher. The positive expression rate of TOP2a in gastric adeno carcinoma was 64.8%(57/88). And TOP2a was negative in the entire normal gastric tissues. The positive expression of TOP2a was not associated with patient’s age (x2=0.008, P=0.927), depth of tumor infiltration(x2=1.856, P=0.173), lymph nodes metastasis (x2=0.030, P=0.863), degree of tumor differentiation (x2=4.404, P=0.111) and clinical stage (x2=0.126,P=0.722). But it was related to the gender (x2=3.928, P=0.047) and distant metastasis (x2=4.368, P=0.037). The results showed that positive rate of TOP2a in male patients was higher than female. The positive rate of TOP2a in patients with distant metastasis was significantly lower than the patients with no distant metastasis. The level of RRM1 and TOP2a in gastric cancer tissue had moderate correlation(r= 0.606,P< 0.05). It had statistical significance. By linear regression analysis, the expression of regression equation was RRM1=3.908+ 0.675TOP2a (r2=0.367, P< 0.05). It had statistical significance.Conclusion:The positive expression of RRM1 was not associated with patient’s age, gender,depth of tumor infiltration depth,lymph nodes metastasis, and distant metastasis,. But it was related to the degree of tumor differentiation, and clinical stage, The stages of gastric cancer was later, the expression of RRM1 in gastric cancer tissues was higher.The stage of tumor was earlier, the expression of RRM1 was lower. The positive expression of TOP2a was not associated with patient’s age, depth of tumor infiltration, lymph nodes metastasis, degree of tumor differentiation and clinical stage. The positive rate of TOP2a in patients with distant metastasis was significantly lower than the patients with no distant metastasis.RRMl and TQP2a paly important role in the process of genetic material duplication and cell division.The level of RRMl and TOP2a in gastric cancer tissue had moderate correlation. It can help to confirm the diagnosis and judge malignant degree of tumor to test the expression of RRMl and TOP2 in gastric cancer tissues. It also has potential significance for selecting individual treatment and estimating prognosis of the patients with gastric cancer. |
