Tuberculosis(TB) is an infectious disease caused by Mycobacterium tuberculosis (Mtb), which is threating human health. In recent years, prevention and treatment of tuberculosis become more and more urgent because of co-infection of TB with HIV and multidrug-resistant TB. Therefore, early diagnosis of the disease remains the most critical importance in TB control. So, it is necessary to identify the specific antigens for TB diagnosis. In this study, we screened and characterized two antigenic complexes for effectively detecting tuberculosis by ELISA analysis.We collected 86 serum samples, including 71 TB patients and 15 healthy individuals, and performed ELISA experiments. Compared with single antigen of Rv1040c, Rv2309A or Rv3872, both antigenic complexes, Rv1040c+Rv2309A and Rv2309A+Rv3872, showed obviously higher sensitivity, specificity and accuracy for TB diagnosis (80.28%, 100%,83.72% for Rv1040c+Rv2309A, and 74.65%,100%,79.07% for Rv2309A+Rv3872).Furthermore, we showed that both antigenic complexes were superior than TB-DOT in terms of sensitivity, specificity, and accuracy (61.97%,73.33%,63.95%), and TB-CHECK-1 in terms of sensitivity, and accuracy(45.07%,54.65%) when comparing with these two commercial kits.Taken together, these two antigenic complexes demonstrated potential application in the ELISA diagnosis of tuberculosis.
