| Objective:The main aim of the study was to observe the efficacy of four Traditional Chinese Medicine Injections(TCMIs, Qingkailing Injection, Shuanghuanglian Injection, Danshen Injection and Dengzhanxixin injection) free of macromolecules,and compare the efficacy with the original injections, in animal experiments.Methods:The four TCMIs free of macromolecules were produced from the original injections with sieves of 10 k at 4℃, 3000 rpm. ⑴ In experiments of Qingkailing Injection and Shuanghuanglian Injection, rabits were randomly divided into seven groups(model group, groups with high, middle, and low dosages of injections free of macromolecules, and groups with high, middle, and low dosages of original injections). Rabit fever model was maked by injecting lipopolysaccharide(1 μg/kg)via auricular vein. Body temperature was measured with ear thermometer, infrared thermal image was recorded, and the animals’ mental state, life signs were also recorded. ⑵ In the experiments of Danshen Injection and Dengzhanxixin injection,rabits were randomly divided into eight groups(blank group, model group, groups with high, middle, and low dosages of injections free of macromolecules, and groups with high, middle, and low dosages of original injections). Blood stasis model was maked by injecting high molecular dextran(T500) via auricular vein. At the end of the experiments, blood sample was drawn, a full set of hemorheology indicators were tested, and animals’ general signs were also recorded.Results:⑴ In experiment of Qingkailing injection or Shuanghuanglian injection, all the rabits’ temperature and general state were normal before model establishment. In model group, temperature increased(about 2-3 degrees than normal) 2 hours later,with apathetic or irritable mind, weakness, hick, and higher frequency of respiration.Five hours later, the rabits’ temperature recovered to normal. Comparison between groups: the difference of animals’ temperatures at the same time was not significant(P>0.05) between original injections and injections free of macromolecules; the difference between model group and original injections and injections free of macromolecules was significant(P<0.05). ⑵ In experiments of Salvia miltiorrhiza Injection or Erigeron Injection, the whole blood viscosity and plasma viscosity of model group were higher at high, middle and low speed, according with the diagnostic criteria of blood stasis(high viscosity blood syndrome). Animals’ general signs: hick, slow-moving, stiffness, dark purple skin mucous membrane and lips, and poor diet were appeared according with high viscosity blood syndrome. The difference of hemorheology was significant(P< 0.05) between blank group and model group; the values of blood flow in groups treated with the two injections(original injections and injections free of macromolecules) were in the normal range, and the values was not significant(P>0.05) between original injections and injections free of macromolecules.Conclusion:The main efficacy of the four injections free of macromolecules was similar to the original. |
