Objective1.To observe the changes of dorsal spinal cord injury(SCI) rat’s micturition upstream pathway by biotin dextran amine (BDA) anterograde neuronal tracing technique in different period.2.To observe the changes of spinal cord injury(SCI) rat’s bladder function,whether the change of the neural pathways associated with bladder function change.MethodsFemale Sprague-Dawley (SD) rats of clean grade 90, were randomly divided into five groups. They are 1 day group,3 days group,7 days group,14 days group and 21 daysgroup.Each group has two sub-group, the control group has 8 amimal,and dorsal spinal cord injury group has 10 animal. We prepared the dorsal spinal cord injury model by ourselves-designed device, and selected T10-L1 as the level of injury. In the control group, the spinal cord was identically exposed, and the spinal cord not damaged. According grouping motor function was evaluated in rats, inclineing muscle tension Ashworth score, Tarlow grade rating, BBB Rating, then each group of rats was measured bladder function and injected BDA into bladder wall at 1 day,3days,7days,21 days after the preparation of animal models espectively.and two weeks after BDA injection, make spinal cord tissues to biopsy and observed. According to the data using homogeneity of variance test results, select the application of Analysis of variance between groups for statistical analysis.Results1. The muscle tension ashworth score of dorsal spinal cord injury(SCI) rats were higher than the control group,with time (p<0.05), showing a reduced gradually returned to normal levels. Compared with the control group, Tarlow grade rating and BBB Rating were lower (p<0.05), with time, Showing increasing tendency gradually returned to normal levels.2. The BDA-positive neurons were stained brown by immunohistochemistry methods. Compared with the control group,we found that the number of BDA-positive neurons of surgical group significantly decreased(p<0.05). Compared with SCI3d and SCI7d group,the numbers of BDA-positive neurons had increased in SCI14d and SCI21d group (p<0.05)3. Urodynamic results showed that no significant difference between the control groups (p<0.05),The bladder maximum capacity of the SCI1d、7d、14d groups were higher than the control groups,but the bladder compliance measurements were lower than the control groups (p<0.05), showing high compliance bladder.Then the bladder maximum capacity of the SCI21d、28d groups were lower than the control groups, but the bladder compliance measurements were higher than the control groups (p<0.05),showing low compliance bladder. The bladder maximum capacity and bladder compliance measurements are significant differences between the SCIld、7d、14d groups and SCI21d、28d groups.Conclusions1. Dorsal spinal cord injury can make the micturition upstream transduction pathway damage. In early stage, it was progressively aggravated. About one week later,the spinal cord nerve cells repaired,and neural pathways could had part of the repair.2. In the shock period of the dorsal spinal cord injury(SCI) rats, the bladder maximum capacity of the SCI1d、7d、14d groups were higher than the control groups,but the bladder compliance measurements were lower than the control groups (p<0.05), showing high compliance bladder. About three weeks after injuryed,the bladder maximum capacity of the SCI21d、28d groups were lower than the control groups, but the bladder compliance measurements were higher than the control groups (p<0.05),showing low compliance bladder.
