Background and ObjectiveOvarian carcinoma is one of the most common gynecological malignancy. In recent years, the incidence of ovarian cancer increased year by year. In various histological types, the most common was epithelial ovarian cancer. The pathogenesis of OC is concealed, non-specific symptoms and short of early screening tool, so most of the ovarian cancer patients presented with advanced stage disease and associate with the highest mortality rate at diagnosis. Early metastasis, recurrence and drug resistance were the characteristics of the ovarian cancer, making ovarian cancer five years survival rates is only 30% ~40%. It is the highest mortality rate in gynecologic oncology cancer, a serious affect on the health of women. However,the mechanism of ovarian cancer remains largely unknown. For the past fewer years CD146 molecule is tumor new vascular endothelial cell marker. It is closely related with the development and metastasis of tumor, angiogenesis and metastasis of cancer cells.Transferrin receptor(transferrin receptor 1, Tf R1) play a key role in the iron homeostasis and regulation of cell growth, which is highly expressed on nucleated erythrocytes and proliferative cell membrane. The latest study showed that CD146 and Tf R1 were closely related with the invasion and metastasis of the malignant melanoma tumor cell. However, the study of CD146 and Tf R1 in the epithelial ovarian cancer has not been reported so far.Therefore, by evaluating the expressions of CD146 and transferrin receptor1(Tf R1) between the cancer,benign and normal ovarian tissues, to investigate their relationship with the EOC genesis, development and metastasis. This study leads a new road to ascertain the possible pathogenesis of ovarian epithelial cancer, and to explore new therapy methods to it.Material and Methods91 cases with epithelial ovarian tumors(including 70 cases with serous adenocarcinoma and 21 cases with mucinous cystadenocarcinoma), 102 cases with benign epithelial ovarian tissues(including 60 cases with serous cystadenoma and 42 cases with mucinous cystadenoma), 54 cases with normal ovarian tissues were to be selected as study objects. Immunohistochemistry was used to detected the expressions of CD146 and Tf R1 in normal,benign and ovarian cancer tissues.Using SPSS 17.0 statistical software for data processing. The differences were considered to be significant with at least P﹤0.05.Results1. CD146 and Tf R1 were detected in both cytoplasm and cellular membrane of the tumor cell. CD146 were also expressed in neovascular endothelial cells.2. The immunohistochemical results showed the positive expression of CD146 gradually increased with the changes of normal,benign and malignant in ovarian epithelial tumor. There were significant differences between all the groups(P﹤0.05). The CD146 expression in the epithelial ovarian cancer was significantly higher than benign ovarian epithelial( χ2=59.171,P﹤0.017). The CD146 expression in the epithelial ovarian cancer was also significantly higher than normal ovarian epithelial(χ2=50.676,P﹤0.017). The CD146 expression of benign epithelial ovarian tissues was higher than normal ovarian epithelia. There differences were not significant(χ2=1.252,P﹤0.017). The Tf R1 expression rate gradually increased with the changes of normal,benign and malignant in ovarian.The differences among these three groups were statistically significant(χ2=77.970,P ﹤ 0.05).Compared with each other among the three groups, and the differences were also statistically significant(all P﹤0.017).3. The advanced the clinical stage, the higher the CD146 and TfR1 expression rate(P ﹤ 0.05).The lower the histopathological differentiation, the higher the CD146 and Tf R1 expression rate(P﹤0.05). The expressions of CD146 and Tf R1 were not correlated with age or pathological patterns(P>0.05).4. According to association analysis, the CD146 and TfR1 expression in ovarian epithelial cancer were a positively correlated(P﹤0.05,rs=0.532).Conclusions1. The CD146 and Tf R1 might contribute to the formation of the ovarian epithelial cancer.2. The high expressions of CD146 and TfR1 might play a key role in the occurrence,progression and metastasis of ovarian epithelial cancer. They also were associated with the degree of ovarian epithelial cancer.3. The CD146 and Tf R1 may synergy in the occurrence, progression and metastasis of ovarian epithelial cancer.
