To study extraction process of Chlorogenic acid、Alkaloids and Flavonids from Root of Ramie. Firstly,we selected the extraction solvent between 70% ethanol solution and water though a pre-test; additionally do some single factor experiments to get the best conditions;finally, we decided a best extraction process of chlorogenic acid、alkaloids and flavonids by the orthogonal experimental. In the purification test, ethyl acetate extraction and polyamide chromatography were used for purifying chlorogenic acid and flavonids of crude extracts from root of ramie; chloroform extraction and D101 macroporous resin adsorption were used for purifying alkaloids. Results show:the best extraction conditions of chlorogenic acid from roots of ramie is that ethanol concentration is 50%, solid to liquid ratio is 1:13, the extraction time is 100 min, the temperature is 90℃, pH is 5. The extraction rate is 0.1202%; and the purity of chlorogenic acid, purifying by ethyl acetate, is 3.34%; yield is 37.14%. After purification with polyamide chromatography, the purity of chlorogenic acid is 12.14%, yield is 17.35%. The best extraction conditions of alkaloids from roots of ramie is that ethanol concentration is 50%, solid to liquid ratio is 1:7, the extraction time is 80 min, the temperature is 90℃, pH is 7. The extraction rate is 0.0844%, and after extraction with chloroform, the five kinds of alkaloid respectively are alkaline phenolic alkaloids, alkaline non-phenolic alkaloids, phenolic tertiary amine alkaloids, non-phenolic alkaloids tertiary amine, strong alkaline water-soluble alkaloids; the purity of Alkaline non-phenolic alkaloids, purifying by D101 macroporous resin, is 6.041%; yield is 4.356%.The results show that the best extraction conditions of Flavonoids in the roots of ramie are as follows:ethanol concentration is 70%, solid to liquid ratio is 1:7, the extraction time is 60 min, temperature is 90℃, and pH is 7. The final purity of Flavonoids is 87.3% after purification.
