Objective:To explore the efficacy and safety of different doses of domestic atorvastatin(ALe)in treatment of acute coronary syndrome(ACS) patients, and to comparative and analysis whether there is difference of different doses of domestic atorvastatin’s clinical adverse reaction, inorder to provide reference basis for future clinical application.Methods:When patients arrived to hospita, the researchers inquired the detailed medical history and complete primary diagnosis,and included them in the experiment,keeped a detailed record of patients’ clinical data in general.The research object were exsanguinated and assayed liver and kidney function,electrolyte,blood sugar,blood fat,myocardial enzymes,troponin I,routine blood and blood coagulation function,and were completed the physical examination and electrocardiogram(ECG)examination. The research object were given the research object normal coronary secondary prevention drugs, such as Aspirin in combination with clopidogrel to antagonize platelet aggregation,nitrates to dilate coronary arteries,beta-blockers to control heart rate and to inhibit myocardial remodeling.The initial research object were received coronary angiography examination toclear the diagnosis of ACS. The patients with serious vascular lesions were gived percutaneous coronary interention(PCI) therapy to improve blood supply situation.The research object were randomly divided into observation group(20mg group,80cases)and the control group(10mg group,80cases).They were informed related situation of experiment, agreed to participate in the experiment, and signed the informed consent form.Regular medication 8 weeks after, the observation target were detected and recorded,which were Total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C), alanine aminotransferase(ALT), aspertate aminotransferase(AST),creatinine(Cr), high-sensitivity c-reactive protein(Hs- CRP) and creatine kinase(CK),etc.The biochemistry analyzer Backman-Au-5821 were unifiedly use to detect for the observation indexes.The adverse drug reactions(ADR) were Closely observed and recorded,such as gastrointestinal discomfort,mental symptoms,myalgia,new diabetes,etc.The statistical analysis for the data from experiment were used by SPSS 16.0 statistical software.Results:Takeing medicine eight weeks after, 2 groups of blood TC were declined dramaticlly, 10 mg group at a rate of 14.3%, 20 mg group at a rate was23.8%.Compared with 10 mg group,20 mg group was more obvious.Twe groups of blood TC were declined dramaticlly,10 mg group at a rate of 16.4%,20 mg group at a rate was 25.7%.Compared with 10 mg group,20 mg group was more obvious.After the medicine,2 groups of angina symptoms were significantly reduced,blood CRP levels basically recovered to normal range.For experimental group(20mg group),the AST/ALT elevation 3 times or more in3 cases,the AST/ALT elevation less than 3 times in 6 cases,gastrointestinal discomfort in 2 cases,myalgia 1 case,serious adverse reactions in total were 6 cases,accounted for7.5%. For control group, the AST/ALT elevation 3 times or more in 2 cases, the AST/ALT elevation less than 3 times in 5 cases,gastrointestinal reaction in 1 case,myalgia in 2 cases, serious adverse reactions in total were 5 cases, accounted for6.4%.The incidence of adverse reactions are similar between the two groups.The differences had no statistical significance(P>0.05).Two groups there were not new diabetes, acute kidney failure, nerve system damage and other adverse reactions. Two groups there were not severe allergic reactions.ConclusionAll in all,10 mg group and 20 mg group both can effectually regulate blood lipid(TC/LDL-C) levels, improve the symptoms of patients with angina pectoris and effectively reduce the blood CRP levels.Early using of large dose ALe(20 mg) has better effect of regulating lipid,and such as rhabdomyolysis, gastrointestinal symptoms and liver function damage and other serious adverse reactions there was no a significant increase in incidence.
