Objectives1.To compare the difference between the insertion of the cartilage perichodrium complex and the self healing time of the ear drum, whether there was a statistical significance;2. To investigate the change of the thickness of the cartilage perichodrium complex in the process of building eardrum by using the cartilage perichodrium complex;3.Observation of the cartilage perichondrium composite for repairing tympanic membrane perforation of tympanic membrane reconstruction, in the different healing periods, reconstruction of the tympanic membrane fiber cartilage film vascular density change and organizational structure and understand significant differences in the number of blood vessels in different periods.MethodsThe research objects are from the animal experimental center of Anhui Provincial Hospital, which is a guinea pig. Forty five healthy male guinea pigs (200-250g) were chosen, all the research object of the feeding, and management of the experimental process are in line with international science and Technology Committee released the "experimental animal management regulations", the Ministry of health released the medical laboratory animal management implementation rules "and Anhui Medical University experimental animal Committee regulations. The experimental guinea pigs for the whole body have no obvious deformity, normal auricle reflex, under ear endoscope examination that the ear canal and tympanic membrane were normal. Will forty-five male guinea pigs were randomly divided into three groups,they were used to create left TM perforation,the left cavum conchae cartilage-perichondrium was immediately used to repair the perforation, the group B (n= five) in guinea pig ear make tympanic membrane perforation modeling, no myringoplasty. Observed time were A (two weeks), group B (six weeks) and group C (twelve weeks), recorded the research object of the tympanic membrane healing time is measured in different healing period.implanted cartilage perichondrium composite of complex thickness, and morphology were observed in the implantation of complex tissue under the microscope, and the micro mirror record fiber cartilage film vascular number of how many. The data is processed by computer SPSS 16.0 analysis software to carry on statistical processing.Results1.Through observation, left ear’s healing time was 3.8+0.84 days, right ear’s time that self healing was 7.2+0.84 days, the healing time of left ear is is shortened obviously than right ear, the difference is significant (p<0.05);2. The thickness of the composite was (group A increased by 14.4%,p<0.05;there was no significant change in thickness of group B after implantion,p>0.05;group C decrease by 14.2%,p<0.05;after implantion,the thickness of cartilage in group C was decreased by 34.2%compared that in group A,p<0.05;3. at the 2nd week in post-implantation, number of vessels within unit area of fiber-perichondrium was up to 13.28±2.49, while the above number reached to 7.71±2.49 at the 6th week after implantation. The final histological structure of reconstructed tympanic membrane at the 2nd week and 6th week was composed of squamous epithelial layer, fibrous-perichondrium layer, cartilaginous layer and mucous layer, while the above histological structure at 12th week was composed of squamous epithelial layer, cartilaginous layer and mucous layer.ConclusionsThe healing time of reconstructed tympanic membrane is shorter than the time spent on healing on its own; the thickness of composite in post-implantation in three groups first increased and then decreased compared to that in pre-implantation; the number of blood vessels of fiber-perichondrium in reconstructed tympanic membrane first increased and then reduced; the layers of final histological structure of reconstructed tympanic membrane are similar to those of normal tympanic membrane.From the perspective of histology, cartilage-perichondrium composite is the suitable material for repairing tympanic membrane.
